摘  要:目的 :观察粉防己碱 (Tet)的抗炎作用及其作用机制。方法 :应用实验性胸膜炎模型观察渗液量和中性粒细胞(Neu)数 ,用福林 -酚法测定蛋白含量 ,用高效液相色谱法测定白三烯B4(LTB4) ,用放射免疫法测定前列腺素E2 (PGE2 )。结果 :在角叉菜胶 (Car)所致大鼠胸膜炎模型 ,Tet剂量依赖性地减少渗液量以及单位渗液的Neu含量 ,但对单位渗液的蛋白含量无明显影响。地塞米松 (Dex)和吲哚美辛 (Ind)也有类似的抗炎作用。Tet和Dex明显地降低LTB4与PGE2 的合成 ,但是Ind仅减少PGE2 生成。结论 :Tet通过抑制Neu游出 ,降低血管通透性发挥抗炎作用 ,它的作用机制与环氧酶抑制剂Ind不同 ,而与Dex相似 ,可能与抑制磷脂酶A2 (PLA2 )AIM The anti-inflammatory action of Tetrandrine(Tet) and the mechanism were investigated in this study.METHODS Experimental pleuritis of rats was used.The neutrophil amount and protein content of the pleural exudate were measured by routine method and Folin-phenol assay respectively.Leukotriene B 4(LTB 4) and prostaglandin E 2(PGE 2) synthesized by Neu were determined by HPLC or RIA.RESULTS In rat pleuritis induced by carrageenan,Tet reduced exudate volume and Neu count per litre in exudate in a dose-dependent manner,but protein content per litre in exudate is not reduced. Dexamethasone(Dex) and indomethacin (Ind) also inhibited inflammatory reaction.Tet and Dex markedly inhibited LTB 4 and PGE 2 synthesis that induced by Neu, but Ind only reduced PGE 2 levels.COCLUSION The anti-inflammatory action of Tet was produced by inhibiting Neu emigration and lowering permeability of capillaries,which is different from cyclooxygenase inhibitor Ind.Tet,like Dex,could producing anti-inflammatory action through inhibiting activity of phospholipase A 2(PLA 2)
