CD44v6在人原发性肝细胞癌的表达及临床意义

摘　　要：目的　观察 CD44v6在人原发性肝细胞癌 (HCC)组织中的免疫组化表达 ,探讨其过量表达与 HCC分型、肿瘤转移及生存期等指标的关系 ,评价 CD44v6可否作为一个新的客观预测 HCC转移及预后的可靠生物学指标 ,方法 :采用鼠抗人 CD44v6单克隆抗体 ,对手术切除的 83例 HCC标本进行了 ABC免疫组化染色 ,以 X2检验、t检验及 log rank检验对结果做统计分析。结果 :CD44v6在 HCC的阳性表达率为 5 3 .0 1% ,CD44v6的阳性表达程度与 HCC转移、肿瘤包膜及预后明显相关 ,但与 HCC原发灶大小、年龄、HBs Ag无关。结论 :CD44v6的表达与 HCC的临床病理生物学行为密切相关 ,特别是Objective Background CD44 is an adhesion molecule involved in cell-to-cell and cell-to-matrix interactions.This transmembrane glycoprotein exists in either standard or variant forms,originated by alternative splicing one of the isoforms (CD44v6)hasbeen shown,in some systems,to modify the metastatic potential of tumor cells.AIM To investigate differential expressions of the human metastasis molecule adhesion CD44 splice variant 6 (CD44v6) in liver carcinomas,and to evaluate whether CD44v6 can be a useful marker in predicting the occurrence,development and metastatic potential of HCC and prognosis of the patients with HCC.Methods 83cases of hepatocellular carcinoma(HCC),Surgically resected specimens were studied immunohistochemically by using a mAb against CD44v6.An antigen retrieval method followed by a routine ABC Process was employed.Result CD44v6 was detected in 53.0% of HCC,The positive staining of CD44v6 was stronger in HCC with tumor metastasis than without tumor metastasis. The patients with positive staining of CD44v6 had a lower postoperative survival rate than those with negative staining.Conclusion The expression of CD44v6 is correlated with the development,metastasis of HCC,as well as with worse prognosis of patients with HCC.CD44v6 is hopefully to be a useful marker for the clinic of HCC.
