摘  要:目的 探讨一氧化碳对缺氧性肺血管结构重建大鼠肺动脉中血小板源性生长因子 (PDGF)表达的影响。方法 将大鼠随机分为 3组 :常氧组 (n =12 )、低氧组 (n =12 )和低氧 +血红素加氧酶组 (低氧 +HO组 ) (n =12 )。以光学显微镜观测 3组大鼠肺中型动脉及小型动脉的相对中膜厚度 (RMT)。应用PDGF的单克隆抗体经免疫组织化学技术对 3组大鼠肺动脉PDGF进行定位及半定量分析。结果 低氧组大鼠中型肺动脉及小型肺动脉RMT均高于常氧组和低氧 +HO组 (P <0 .0 1)。 3组大鼠各级肺动脉内皮细胞中均有PDGF的表达 ,低氧组高于常氧组和低氧 +HO组 (P <0 .0 1)。中型肺动脉内皮细胞中PDGF的表达与相应中型肺动脉RMT之间呈正相关(r为 0 .82 3 5,P <0 .0 1) ;小型肺动脉内皮细胞中PDGF的表达与相应的小型肺动脉RMT之间呈正相关 (r为0 8642 ,P <0 .0 1)。Objective To investigate th e impact of carbon monoxide (CO) on the expression of platelet derived growth factor (PDG F) in the mechanism regulating hypoxic pulmonary vascular remodeling.?Methods 36 rats were randomly divided into 3 groups ( n =12 each ): normoxic, hypoxic and hypoxic + heme oxygenase (hypoxic+HO). The relative med ial thickness (RMT) of small and medium muscularized pulmonary arteries was dete rmined under light microscope. Immunohistochemical technique was used to assess the abundance and localization of PDGF in pulmonary arteries.?Result s The RMT of small and medium pulmonary arteries in hypoxic rat gr oup was markedly higher than that in the normoxic group and hypoxic+HO group( P <0.01). PDGF was expressed in smooth muscle cells of pulmonary artery in all of the 3 groups,more marked in the hypoxic group than in the other two( P < 0.01). There was a positive correlation between RMT and PDGF expression in medi um and small pulmonary arteries ( r =0.8235 and 0.8642 respectively, P <0.01 ).?Conclusion Carbon monoxide inhibits the PDGF expressi on in pulmonary arteries of rats undergoing hypoxic pulmonary vascular remodelin g.
