儿童横纹肌肉瘤中p16和pRb蛋白的表达及其意义

摘　　要：目的探讨横纹肌肉瘤(RMS)中p16和pRb的蛋白表达及其与RMS的发生、浸润和转移的关系。方法应用免疫组化方法检测30例RMS病人中p16和pRb蛋白的表达情况。结果30例RMS中,无1例p16和pRb两种蛋白表达均正常。9例p16阴性的RMS中,有4例pRb呈强阳性表达;4例p16弱阳性病例,pRb均呈强阳性;17例p16中-强阳性病例中,有3例pRb阴性表达,12例弱阳性表达。而5例pRb阴性RMS中,3例p16强阳性表达。在21例p16蛋白阳性的病例中,有5例发生转移,而在p16蛋白阴性的9例中有7例发生转移。在p16阴性和阳性组之间P=0.0125,有显著性差异。pRb阳性的25例中,有10例发生转移,在pRb阴性的5例中有2例转移。在pRb阴性和阳性组之间,P=1.0000,无显著性差异。结论(1)p16、pRb异常与RMS的发生密切相关;(2)p16与pRb表达呈负相关关系;(3)p16的表达缺失与RMS的广泛浸润和转移有关。Objective To evaluate the significance of p16and pRb expression in children rhabdomyosarcoma(RMS)and the relationship between the two proteins.Methods 30cases of RMS were studied for p16and pRb expression with immunohistochemical method.Results None of all the cases was both p16and pRb normal in the two proteins.In the9cases of RMS negative to p16,4cases were strongly positive.In the4cases of RMS faintly positive to p16,all of the4cases were strongly positive.In the17cases of RMS moderately or strongly positive to p16,3cases were negative,12cases faintly positive.In the5cases of RMS negative to pRb,3cases were strongly positive.In the21cases of RMS positive to p16,5cases had invasive growth or metastasis.In the9cases of RMS negative to p16,7cases had invasion or metastasis.Between p16negative and positive groups had significant difference (P=0.0125).In the25cases of RMS positive to pRb,10cases had invasion or metastasis.In the5cases RMS negative to pRb,2cases had invasion or metastasis.Between pRb negative and positive groups,P=1.0000,don't have significant difference.Conclusions (1)The abnormality of p16and pRb are highly related to the pathogenesis of RMS.(2)There is a negative reciprocity between p16and pRb protein expression in RMS.(3)Loss of p16ex pression is related to invasion or metastasis.
