摘 要:具有拮抗大白菜软腐病功能的菌株 90 2 1 2 (Pseudomonas.sp .)的培养及最大菌落数产生的发酵模拟生产工艺条件 :经正交试验得到最适的培养基碳、氮源组成 (% )为 :葡萄糖 1 .5 ,大豆饼粉 2 .0 ,淀粉 1 .0 ,酵母粉 1 .5 ;最适生长温度为 30℃ ;pH7.0 ,接种量 4 % ,在 2 5 0mL三角瓶装 70mL培养基 ,2 0 0r/min摇床上培养 35h ,菌落数为 1 .2 5CFU× 1 0 9;发酵罐发酵 (1t罐 ) 2 2h ,终点总菌落数为 1 .4 8× 1 0 1 0 CFU/mL。由上述工艺制得的该生防制剂软腐灵 ,经田间试验结果表明 :对软腐病菌有强烈的抑制作用 ,大白菜用软腐灵处理二次的防病增产效果优于只用一次的处理 ,对大白菜软腐病有 70 %~ 80 %的防治效果 ,并有 2 0It was found that strain 90 2 1 2 (Pseudomonas) could control soft rot of Chinese cabbage. The simulated conditions of producing strain 90 2 1 2 were studied through different culture steps and orthogonal test of different medium compositions. The results showed that the optimum medium composition was: glucose (1.5%), soybean meal (2.0%), starch (1.0%),and yeast powder(1.5%).The optimum growth temperature was 30℃.When cultured for 35 hours on a 200r/min shaker with 70mL of medium in 250mL flask at pH7.0 and 4% pitching rate, the total bacterial count was 1.25×10 9CFU/mL. When the fermentation in 1 ton fermenter for 22 hours was made,the final total bacterial count was 1.48×10 10 CFU/mL. The results from field experiment indicated that the preparation Ruanfuling made through above technology had effects on controlling soft rot of Chinese cabbage and increasing the crop yield, and the effect of applying Ruanfuling to the crop twice was better than that of doing once.The control efficiency was 70%~80% and the rate of increasing the crop yield was about 20%.
