胶质瘤组织明胶酶B基因表达研究

摘　　要：目的 了解明胶酶B在胶质细胞瘤中的作用。方法 36例胶质细胞瘤,其中Ⅰ～Ⅱ级胶质瘤13例,Ⅲ～Ⅳ级胶质瘤23例。患者术中取肿瘤组织及正常脑组织,提取组织总RNA;用逆转录共扩增定量PCR方法测定明胶酶B(MMP_9)基因表达水平。结果 Ⅲ～Ⅳ级胶质瘤明胶酶B基因表达水平(1.77±0.55)明显高于Ⅰ～Ⅱ级胶质瘤(0.63±0.11)及正常脑组织(0.46±0.08)。胶质瘤MMP_9基因表达水平与肿瘤大小无关,而与肿瘤组织分化程度有关。结论 明胶酶B在脑胶质瘤的进展中可能起重要作用。Objective To investigate the role of extracellular matrix degradation in glioma.Methods 23 patients with grade Ⅲ -Ⅳ glioma and 13 patients with grade Ⅰ - Ⅱ glioma were selected and normal brain tissues were obtained during operation. Total RNA was isolated and gelatinase B ( MMP9) gene expression level was measured by reverse transcription polymerase chain reaction. Results The gelatinase B (MMP9 ) gene expression level in grade Ⅲ-Ⅳ glioma tissue( 1.77 ± 0.55) was much higher than that in grade Ⅲ-Ⅳ glioma(0.63 ± 0.11) and normal brain tissue(0.46 ± 0.08). MMP9 gene expression level had strong correlation with the grade of histological diferentiation and no correlation with tumor size. Conclusion Gelatinase B may play an important role in the progression of glioma.
