摘　　要：目的　探讨脑星形细胞瘤中CDK4基因蛋白的表达 ,及其与星形细胞瘤病理学分级关系。探讨G1→S期CDK4在星形细胞瘤发生 ,发展过程中的作用。方法　采用SP免疫组化技术结合高温高压抗原修复法 ,在光学显微镜下检测 33例人脑星形细胞瘤中CDK4蛋白的定位、定量及其阳性表达率与病理分级之间关系。结果　星形细胞瘤细胞中CDK4基因的表达高于正常对照脑组织细胞 ,CDK4阳性表达主要位于胞浆。CDK4阳性表达率在各级别间比较有显著差异性差异 (P <0 .0 5 ) ,同时发现低级别肿瘤中有 3例CDK4基因呈过度表达 ,占 15 % ,而在高恶性度组星形细胞瘤中均存在CDK4基因过度表达。结论　在脑星形胶质细胞瘤中存在着CDK4基因蛋白的异常表达 ,其与脑星形细胞瘤的发生及恶性发展可能有关 ,CDK4基因的异常表达对星形细胞瘤病理分级有指导意义。Objective To discuss the expression of CDK4 gene in astrocytoma , and the relationship between it and pathological classification. To discuss the CDK4 gene proteins' functions among the astrocytomas' occurence and development.Methods A S-P immunohistrochemical technique is undertaken to detect the location, the quantity and the positive expression rate of CDK4 gene in 33 human brain astrocytomas.Results The CDK4 gene's expression in Astrocytomas groups are all higher than in nomal control group. CDK4 is mainly located in cell cytoplasm. There exist CDK4 gene abnormal expression in astrocytomas, the positive rate of CDK4 gene exist remarkable difference between different grade (P<0.05), besides there exist CDK4 overexpression in 3 low-grade gliomas while exist in all the high-grade gliomas.Conclusion there exist overexpression of CDK4 gene in astrocytoma,the overexpression have relation with the astrocytoma's occurrence and development, the overexpression of CDK4 can direct the pathological grade.
