摘 要:研究了有机相中脂肪酶催化α 氰基 3 苯氧基苄醇乙酯的醇解化反应 ,制备α 氰基 3 苯氧基苄醇 .考察了酶、溶剂、醇、醇用量、溶剂水含量以及底物浓度等因素对反应的影响 ,结果表明Novozym43 5脂肪酶催化活性最高 ,经实验确定的最佳条件为 :脱水甲苯为溶剂 ,正辛醇为酰基受体 ,正辛醇∶酯的摩尔比为 1 5∶1,酶量为 8mg/mL时的最佳底物浓度为 10 8 13mmol/L ,在上述条件下反应 3 0h酯的转化率 >96% .The lipase-catalyzed alcoholysis of α-cyano-3-phenoxybenzyl acetate (CPBAc) in organic solvents for the synthesis of α-cyano-3-phenoxybenzyl alcohol (CPBA) was studied. The experimental results indicated that Novozym435 lipase showed the highest catalytic activity for the reaction. 1-Octanol was founed to be the best acyl acceptor. Toluene was the preferred solvent and the optimum molar ratio of 1-octanol to CPBAc was about 1.5∶1. The reaction rate was slowed down with the addition of water to the solvent. The best concentration of substrate was 108.13 mmol/L under 8 mg/mL lipase for the reaction. When the reaction was carried out under the above optimum conditions, more than 96% of conversion was achieved.
