摘  要:目的 :观察雌激素受体α(estrogenreceptorα,ERα)免疫阳性星形胶质细胞在阿尔茨海默病 (Alzheimerdisease,AD)病人组和老年对照组海马中的分布变化。 方法 :采用免疫荧光细胞化学双标方法和激光共聚焦扫描显微镜。结果 :ERα免疫阳性星形胶质细胞主要分布在齿状回的门和海马的分子层、放射层及始层。GFAP (glialfibrillaryacidicprotein)免疫阳性细胞、ERα胞核染色的和ERα胞质染色的星形胶质细胞 (number/mm2 )在AD组的CA1均显著增加 (P分别小于 0 .0 0 1,0 .0 5 ,0 .0 5 ) ,但其ERα核着色或质着色的星形胶质细胞占GFAP免疫阳性星形胶质细胞的百分数 ,在对照组 (2 .5 6 %,n =13;16 .6 8%,n =13)和AD病人组 (2 .15 %,n =13;2 6 .0 0 %,n=13)之间无显著差异 (P >0 .0 5 )。结论 :这些结果表明 ,在AD的发病过程中雌激素可能通过ERα介导对海马星形胶质细胞的作用。Objective: To investigate the distribution and alteration of estrogen receptor α (ERα)-immunoreactive astrocytes in the hippocampus of control and Alzheimer disease (AD). Methods: Means of fluorescence immunocytochemistry was applied. Results: ERα-immunoreactive astrocytes were mainly located in the hilus of the dentate gyrus and the stratum moleculare,stratum radiatum,and stratum oriens of the hippocampus. The number of GFAP (glial fibrillary acidic protein)-immunoreactive astrocytes,nuclear ERα-staining astrocytes,and cytoplasmic ERα-staining astrocytes per unit area (1 mm 2 ) significantly increased ( P <0.001,P <0.05,P <0.05,respectively) in CA1 in AD patients,while the percentage of ERα-immunoreactive astrocytes in GFAP-immunoreactive astrocytes between the two groups did not differ significantly ( P >0.05). Conclusion: The data suggest an important role for ERα-mediated effects of estrogen on astrocytes in the hippocampus of AD patients.
