摘  要:用高效液相色谱法测定从(木奈)果实中纯化提取的3种植物内源激素GA_3、IAA.ABA(±),并介绍了植物组织中激素日常分析的提取和纯化过程。本实验以C_(18)柱为分离柱;甲醇:0.01mol/L,H_3PO_4(pH2.4)=42:58(v/v)为流动相,在流速1mL/min时恒流洗脱,λ=254nm处检测了3种内源激素。被测组分的含量与其峰面积有良好的线性关系(r=0.9996~1.000),加标回收率在92.13%~95.25%,同时文中还评估了不同贮藏温度和不同试剂处理对(木奈)果实中GA_3含量的影响。3种内源激素的最小检出限均在纳克级。This parper describes the application of reversed-phase HPLC to the determination of internal plant hormoned [gibberellin (GA3), indole-3-acetic acid (IAA) and abscisic (ABA)] in purified extracts of Nane fruit, and also gives an extraction and purification procedure useful for the routine analysis of the hormones in plant tissue. A Cu column was used and isocratic elution with MeOH: 0. 01 mol/L H3PO4 (pH 2. 4) =42 :96-1. 000. Meanwhile, the affect of various storage temperatures and reagent treatments on the content of GA3 in Nane fruit is examined in the paper. The lowest detecable limit for three hormones were in nanogram le 58 (v/v) at a flow rate of 1 mL/min, was performed with a UV monitor at 254nm. The average recovery of four internal plant hormones were 92.13%-95. 25% (n = 5) and correlation coefficients were 0. 99vel.
