表达狂犬病病毒糖蛋白的重组鸡痘病毒的构建和鉴定

摘　　要：将 RVG基因插入到鸡痘病毒表达载体 p UTA- 2 Sm a 位点获得重组转移载体 p U TA- RVG,利用脂质体转染已感染中国鸡痘病毒疫苗株 2 82 E4株 2～ 3h的鸡胚成纤维 (CEF)细胞 ,收获病毒后 ,用 Brd U法进行加压筛选重组病毒 ,PCR检测到重组病毒 RVG基因 ,Western blot检测出 RVG,从而证实已成功构建了表达RVG gene was cloned into the SmaⅠ site of the FPV expression vector pUTA-2 to construct the recombinant transferring plasmid pUTA-RVG.Chicken embryo fibroblast(CEF) cell cultures which had been infected with fowlpox virus(FPV) chinese vaccine strain 282E4 for 2-3 hours were transfected with pUTA-RVG plasmid DNA by liposomal transfection.Recombinant FPV were screen out under the pressure of the BrdU.In the selected recombinant FPV the RVG gene was detected by PCR and the RVG by western blot,so the recombinant FPV expressing the RVG was successfully constructed.
