摘 要:目的 研究四氢喋呤 (BH4 )对兔实验性缺血再灌注 (MI R)损伤中一氧化氮 (NO)、丙二醛 (MDA)、超氧化物歧化酶 (SOD)水平及心肌梗死面积的影响。方法 2 4只新西兰兔随机分为假手术组、模型组和药物干预组 3组。制备MI R模型 ,观察BH4 预处理后MI R损伤中NO、MDA、SOD水平的变化及对心肌梗死面积的影响。结果 模型组较假手术组NO显著减低 [(2 8± 10 ) μmol L比(116± 17) μmol L ,P <0 0 1],MDA水平明显升高 [(5 8 3± 10 4 )nmol L比 (7 0± 1 8)nmol L ,P <0 0 1],SOD活性明显减低 [(2 6 8± 17)nu ml比 (340± 2 4 )nu ml,P <0 0 5 ];而BH4 预处理组较模型组NO明显升高 [(6 2± 17) μmol L比 (2 8± 10 ) μmol L ,P <0 0 1],MDA明显减低 [(30 9± 6 1)nmol L比(5 8 3± 10 4 )nmol L ,P <0 0 1],SOD活性无明显改变 [(2 88± 2 0 )nu ml比 (2 6 8± 17)nu ml,P >0 0 5 ];BH4 预处理组和模型组心肌梗死面积差异有显著性 [(18± 4 ) %比 (16± 4 ) % ,P <0 0 5 )。结论 MI R导致内皮功能紊乱。BH4 对内皮功能有保护作用 ,从而减轻再灌注损伤。Objective To evaluate the influence of tetrahydrobiopterin on NO and MDA concentration and SOD activity as well as the myocardial infarction area. Methods Twenty-four rabbits were divided into three groups randomly: control group,MI/R group and BH 4 group. After occlusion of the left anterior descending coronary artery of a rabbit for 60 minutes followed by 90 minutes of reperfusion (ischemia/reperfusion),NO and MDA and SOD concentrations were measured and the myocardial infarction area was caculated. Results Compared with control group,the NO concentration in MI/R group was markedly decreased [(28±10) μmol/L vs (116±17) μmol/L,P <0.01],the MDA concentration was markedly increased [(58.3±10.4) nmol/L vs (7.0±1.8) nmol/L,P <0.01],and SOD activity was markedly decreased [(268±17) nu/ml vs (340±24) nu/ml,P <0.05]: Compared with MI/R group,the NO concentration in BH 4 group was markedly increased [(62±17) μmol/L vs (28±10) μmol/L, P <0.01] and the MDA concentration was markedly decreased [(30.9±6.1) nmol/L vs (58.3±10.4) μmol/L,P <0.01]: There were significant difference between MI/R and BH 4 groups with SOD activity [(288±20) nu/ml vs (268±17) nu/ml,P >0.05];There was significant difference between MI/R and BH 4 groups in infarction area,too. [(18±4)% vs (16±4)%,P <0.05]. Conclusion Reperfusion injury results in the endotheliai dysfunction,and tetrahydrobiopterin restores endothelial function in rabbits after reperfusion injury.
