MUC2在结肠癌及结肠腺瘤的表达及意义

摘　　要：目的探讨粘蛋白2(Mucin2,MUC2)在结肠癌与结肠腺瘤的表达及与临床病理参数之间的关系。方法选取经病理证实的电子肠镜活检的结肠正常组织、结肠腺瘤组织及外科手术切除的结肠癌组织的石蜡包埋标本(结肠正常组织25例、结肠腺瘤组织25例、结肠癌组织25例),应用免疫组化方法检测MUC2的表达。应用Image-pro Plus(IPP)软件分析免疫组化图片,测量阳性区域平均光密度值(average optical density,AOD),根据AOD值的大小判断MUC2的表达强度。结果 1MUC2在正常对照组结肠组织中的表达AOD值为(0.0665±0.0252),结肠腺瘤组AOD值(0.0476±0.0230),结肠癌组AOD值(0.0330±0.0280),表达强度逐渐降低,各组间比较差异有统计学意义(P均<0.05)。2在结肠癌组织中,MUC2的表达与淋巴转移、Dukes分期及肝转移(P=0.019、0.019、0.029)均呈负相关,而与患者性别、年龄、肿瘤部位及肿瘤分化程度无相关(P=0.807、0.689、0.206、0.779)。结论 MUC2可能参与结肠癌的发生、发展,可能是结肠腺瘤癌变的潜在标志物。Objective To investigate the expression and clinical significance of MUC2 in colon carcinoma and colonic adenoma. Methods Immunohistochemical staining was performed to detect the pathologically confirmed electronic enteroscope biopsy and the expression of MUC2 in 25 of colon normal tissues,25 of colon adenoma and 25 of colon carcinoma which were resected by surgical operation. The immunohistochemical pictures were analyzed and the positive regional average optical density( AOD) was measured the Image- pro Plus( IPP) software. The expression intensity of MUC2 was judged based on the size of the AOD values.Results The expression of MUC2 gradually and significantly reduced in normal colon tissue,colon adenomas,colon cancer tissue( P < 0. 05); The expression level of MUC2 had significantly negative connection with lymph node metastasis,Dukes stage and liver metastasis( P = 0. 019,0. 019,0. 029) in colon cancer but not with gender,age,tumor location and tumor differentiation degree( P = 0. 807,0. 689,0. 206,0. 779).Conclusion The down- regulation of MUC2 expression involved in carcinogenes and progression in colon cancer and it may be the potential markers to colon cancer.
