胃癌BGC-823细胞增殖与NOTCH1和HES1表达的关系

摘　　要：目的探讨HES1蛋白在胃癌细胞增殖中的作用。方法在5%CO2、37℃、饱和湿度条件下培养胃癌BGC-823细胞和肝干细胞系(LEPC),MTT法检测细胞增殖活性,免疫细胞化学检测HES1和NOTCH1表达。结果培养48h时,BGC-823和LEPC细胞生长良好,MTT比色法检测显示,BGC-823细胞组增殖能力明显强于LEPC组,BGC-823组培养24、48和72h的OD值(分别为0.67±0.17、1.02±0.14和1.22±0.24)均明显高于LEPC组(分别为0.36±0.16、0.35±0.07和0.38±0.14)(P均<0.05)。免疫细胞化学染色可见,少数BGC-823细胞核HES1阳性,比较多的BGC-823细胞核周胞质呈HES1阳性,少数细胞阳性着色见于细胞质和细胞核;HES1阳性以弥漫性表达为主,细胞质表达明显多于细胞核,巨细胞表达更加明显。LEPC组部分细胞HES1免疫组化染色阳性,阳性着色主要见于细胞核。在培养48和72h,BGC-823组HES1蛋白免疫细胞化学染色平均光密度值(OD)均明显高于LEPC组(P<0.05)。BGC-823大多数细胞NOTCH1阴性,少数阳性。LEPC组可见部分细胞NOTCH1免疫组化染色阳性。在培养24、48和72h,BGC-823组NOTCH1平均OD值与LEPC组的差异无统计学意义(P>0.05)。结论 HES1高表达与BGC-823细胞增殖能力较强有关。Objective To investigate the effect of HES1 on the proliferation of gastric carcinoma cell. Methods The gastric carcinoma cell line BGC-823 and Liver epithelial progenitor cells( LEPC) were cultured under 5% CO2 and 37℃. The level of cell proliferation was measured with MTT. The expression of HES1 and NOTCH1 were observed by immunocytochemistry. Results The good profile of BGC-823 and LEPC was demonstrated at 48 h. As MTT test showed,the cell proliferation of BGC-823 was significantly higher than that in LEPC group( P < 0. 05). At 24 h,48h,and 72 h,the optical densities( OD) in BGC-823 group( 0. 67 ± 0. 17,1. 02 ± 0. 14,and 1. 22 ± 0. 24) were significantly higher than those in LEPC group,0. 36 ±0. 16,0. 35 ± 0. 07 and 0. 38 ± 0. 14( P < 0. 05). The positive staining of HES1 was observed in the nucleus of a few cells,the cytoplasm of most cells,and both,nucleus and cytoplasm,of few cells at BGC-823 group and the more immunostaining was showed in the giant cells. The immunostaining of HES1 was observed in a few cells at LEPC group and the positive signal was showed in the nucleus. At 48 h,and 72 h,OD of HES1 immunostaining in BGC-823 group were significantly higher than that in LEPC group( P < 0. 05). The immunostaining of NOTCH1 was observed in a few cells at BBGC-823 group and the most cells was negative stained. The immunostaining of NOTCH1 was observed in a few cells at LEPC group. At 24 h,48h,and 72 h,OD of NOTCH1 immunostaining in BGC-823 group were similar to that in LEPC group( P > 0. 05). Conclusion The over-expression of HES1 was involved in the proliferation of gastric carcinoma cell line BGC-823.
