摘　　要：目的 :检测TNP— 4 70对体外培养人口腔鳞癌颈淋巴结转移癌细胞系GNM的抑制作用。方法 :利用台盼蓝排斥法、生长曲线测定法及MTT法检测不同浓度TNP— 4 70对GNM细胞的抑制作用。结果 :1) 0 .0 5～ 2 5 μg·mL-1的TNP— 4 70对GNM细胞的生长有不同程度的抑制作用 ,抑制率与药物浓度成正相关 ,TNP— 4 70的半数抑制浓度IC50为 0 .5 μg·mL-1。 2 )TNP— 4 70与平阳霉素合用 ,对GNM细胞的抑制率较单用TNP— 4 70或平阳霉素提高。结论 :TNP— 4 70可抑制GNM细胞的生长 。Objective:To detect the suppression of TNP—470 on neck lymph node metastasis GNM cell line derived from human oral squamous cell carcinoma in vitro.Methods:Dyeing exclusion method,growth curve assay and MTT assay were used to detect the suppressive effect of different concentration TNP—470 on GNM cell line.Results:1)Different suppressive effect were observed when 0.05～25 μg·mL -1 TNP—470 were added on GNM cell line,and the inhibitory rates were correlated with the concentration of TNP—470 (50% inhibitory concentration,approximately 0.5 μg·mL -1). 2)When TNP—470 was combined with Pingyangmycin,a synergetic effect on GNM cell line was observed.Conclusions:TNP—470 can significantly inhibit GNM cell line growth,and this effect shows apparently dosedependent.
