抗大肠杆菌IgY对大鼠全腹照射后肠源性感染的预防作用

摘  要:目的 建立大鼠全腹照射模型 ,观察抗大肠杆菌IgY管饲对大鼠肠粘膜结构的变化和细菌移位的影响及对肠源性感染的预防作用 ,为临床上预防放射性肠粘膜损害以及肠源性感染提供实验依据。方法 健康SD大鼠 30只 ,体重 ( 2 0 0± 2 0 ) g ,雌雄各半。大鼠随机分为 3组 (每组 10只 ) :正常对照组 (A组 )、照射对照组 (B组 )、IgY预防组 (C组 )。用直线加速器对大鼠进行全腹照射 ,总照射剂量为 10 0 0cGy。自照射前一天开始进行IgY管饲。A、B两组大鼠不灌任何药物 ,C组大鼠以 10 %抗大肠杆菌IgY水溶液 1.5ml管饲 ,q12h。持续 4d。照射后第 4天处死大鼠 ,无菌条件下打开腹腔 ,取肝、脾及肠系膜淋巴结制成匀浆 ,行细菌培养 ,统计细菌移位率 ;从心腔采血 ,分离血清 ,测定内毒素水平。结果 B组细菌移位明显 ,肝、脾、淋巴结组织细菌培养总阳性率达 96 .7% ,血清内毒素水平明显升高 ;C组肝、脾、淋巴结组织细菌培养总阳性率为 6 .7% ,血清内毒素水平下降 ,且与A组相比均有显著性差异 (P <0 .0 5~ 0 .0 0 1)。结论 抗大肠杆菌IgY能明显降低全腹照射后肝、脾、肠系膜淋巴结的细菌培养阳性率 ,提示具有保护大鼠肠粘膜屏障 ,减少细菌移位的作用 ;并且能改善反映大鼠肠源性感染的指标 ,降低血中内毒素 ,提示对肠源性?Objective In order to observe bacterial translocation and prevention of intestinal mucosa damage and gut origin septic states in animal model of abdominal radiation by IgY.WTHZ]Methods Methods Thirty health rats weight 200±20 gram,female and male each half.Rats were randomly divided into three groups:normal control group A,group B radiated merely and group C received IgY.Rats were abdominal radiation by linear accerlerator,total dose was 1000cGy.IgY were poured before the day of radiation.group C poured 10% anti colibacillus IgY 1.5 ml twice a day for four days.On the forth day after radiation,rats were anesthesia by Ketamine and killed on sterile operation table.In strict sterile condition , open abdominal cavity,a piece of liver,spleen and two pieces of mesenteric lymph node were removed and homogenized,then cultured.Bacterial translocation incidence was evaluated.At the same time,blood was withdrawal from heart,segregated serum,preserved at -20°C.Endotoxin serum was assayed.Results In group B,bacterial translocation was obviously,total bacterial translocation incidence of liver,spleen and mesenteric lymph node was 96.7%;the content of serum endotoxin increased.Group C total bacterial translocation incidence of liver,spleen and mesenteric lymph node was 6.7%.The content of serum endotoxin decreased.There was satistically significant difference between group C and group B ( P <0.05~0.001).Conclusion Anti colibacillus IgY can decrease bacterial translocation incidence of liver,spleen and mesenteric lymph node after abdominal radiation obviously.It indicated that IgY can protect rat intestine mucosa barrier,reduce bacterial translocation incidence.Anti colibacillus IgY improved the serum indexs reflecting gut origin septic states,decrease the content of endotoxin recovered to normal. It indicated that IgY can prevent gut origin septic states.
