硒代二半胱氨酸对白血病细胞系的诱导分化研究

摘  要:硒抑制体外白血病细胞的生长和增殖,并能诱导部分白血病细胞分化成熟,但其药理作用因硒制剂的不同而异.硒代二半胱氨酸抑制V937和K562白血病细胞的生长和增殖,半数抑制浓度为3.0μmol/L.经3.0μmol/L硒代二半胱氨酸作用3天后,U937,细胞吞噬率以5%上升至14%,K562细胞内血红蛋白含量由0.20增至0.40μg/106细胞,说明硒代二半胱氨酸能诱导部分白血病细胞分化成熟.The selenium compound-dicysteine selenium has been shown to inhibit the growth of U937 and K562 leukemic cells with IC50 of 3. oμmol/L. Furthermore.the differentiation-inducing effects were observed. After incubation for 3 days with dicysteine selenium at concentration of 3. oμmol/L. morphological and functional differentiation has occurred in some of the leukemic cells. 14% U937 cells were capable of phagocytosis after treated with dicysteine selenium while only 5% U937 cells were capable of phagocytosis in controls.Meanwhile dicysteine selenium was effective in stimulating hemoglobin synthesis in K562 cells because more hemoglobin product was determined in drug-treated cells(0. 4μg/106 cells) than in controls (0.2μg/106 cells).
