家蝇血淋巴的提取及抗菌物质的诱导

摘　　要：本文报道了大肠杆菌注射和超声波处理家蝇(Musca domestica)后对家蝇抗菌物质的诱导效果,抗菌物质的活性,抗菌的专一性,并研究了家蝇血淋巴的提取方法。结果表明,通过超声波法和注射法均可诱导家蝇产生抗菌物质,其活力高峰出现在诱导后60小时。诱导后48小时的血淋巴稀释100倍,对大肠杆菌的溶菌活力达0.115单位,其抗菌物质对大肠秆菌(Escherichia coli)、金黄色葡萄球菌(Staphlococcus nureus)、大白菜软腐病菌(Erwiniacarotovora)、黑腐病黄色单胞菌(Xanthomonas campestris)苏云金杆菌(Bacillus thuringie-nsis)等均有抑菌作用,而对真菌白僵菌(Beauveria bassiana)未发现抑菌作用。对幼虫的诱导效果好于成虫。血淋巴的提取方法由以往的毛细管吸取法改为离心的方法使产量提高2倍,工作效率提高50倍。Antibacterial matter was induced from house fly(Musca domestica)by injecting E.coilinto its body and by using sonic treatment.The activity and specificity of antibacterial mat-ter were reported and the abstraction method of hemolymph was also described.The resultsshows that both injection and sonic method were able to induce antibacterial matter from hou-se fly and the activity peak value appeared across 60 hours after treatment.After 48 hoursand at the dilution of 100 times,the activity of antibacterial matter in hemolymph is 0.115unit.The antibacterial matter could also have restriction effect on the growth of E.coli,Sta-phlococcus aureus,Erwinia carotovora,Xanthornonas campestris,and Bacillus thuring(?)ensisbut it could not act on Beauveria bassiana.Inducing effect of the antibacterial matter on larvais higher than on adult insect.By using centrifuge method the yield of hemolymph is 2 timesthan capillary method used.The efficiency of centrifugal method is 50 times that of the capil-lary method.
