端粒长度在胃癌组织中的改变及临床意义

摘　　要：目的探讨端粒长度的变化与胃癌形成的关系。方法采集相对正常胃黏膜10例、癌前病变15例、胃癌20例、淋巴结转移癌组织5例;Southern blot分析端粒长度的变化。实验数据采用方差分析,P<0.05有统计学意义。结果正常胃黏膜、癌前病变、胃癌及淋巴结转移癌组织平均端粒长度分别为(9.20±1.09)、(8.70±0.81)、(7.66±1.77)及(7.30±0.42)kb。方差分析癌前病变组、胃癌组和转移癌组的端粒长度比正常组短;转移癌组、胃癌组的端粒长度比癌前病变组短(P<0.01)。胃癌中端粒长度与患者的年龄、性别、肿瘤大小、肿瘤的分化程度和临床分期无关(P>0.05)。结论正常胃黏膜、癌前病变、胃癌与转移癌组织的平均端粒长度逐渐缩短。端粒长度的改变可能参与胃癌形成过程,端粒缩短可能是胃癌发生的早期事件。Objective To explore the associations between the changes of telomere length and human multistage carcinogenesis of gastric cancer.Methods The sample included 10 normal gastric mucosa,15 precancerous lesions,20 gastric cancer(GC) and 5 lymph nodes with gastric cancer metastasis(GCLM).The mean telomere length was measured using Southern blotting method.ANOVA was used to compare among different groups,and P<0.05 was considered as statistically significant.Results The mean(SD) of the telomere length was 9.20(1.09),8.70(0.81),7.66(1.77),and 7.30(0.42) respectively for normal gastric mucosa,precancerous lesions,GC,and GCLM.Results of ANOVA showed the telomere lengths were shorter in precancerous lesions,GC,and GCLM compared to those in normal gastric mucosa tissues(P<0.05).The telomere lengths in GC and GCLM were significantly shorter than those in precancerous lesions(P<0.05).The telomere lengths were not related to patients' age,sex,caner size,cancer differentiation,and the clinical stages of cancer(P>0.05).Conclusions The mean telomere lengths in the normal gastric mucosa is the longest among all of these patients,and followed by precancerous lesions,GC,and GCLM.Telomere length may be involved in the carcinogenesis and progression of gastric cancer.A short telomere could be an early event of gastric cancer.
