胃癌中HER2蛋白表达及基因扩增的临床意义

摘　　要：目的探讨类表皮生长因子受体2(human epidermalgrowth factor receptor 2,HER2)蛋白表达及基因扩增与胃癌临床病理特征的关系。方法分别用免疫组织化学方法和荧光原位杂交方法检测60例胃癌与癌旁非肿瘤胃黏膜中HER2蛋白表达和基因扩增状态,分析HER2蛋白表达及基因扩增与胃癌各临床病理特征的相关性。采用SPSS 16.0统计软件对数据进行x^2检验分析,P<0.05为差异有统计学意义。结果 60例癌旁非肿瘤胃黏膜中无HER2蛋白表达及基因扩增。胃癌组织中HRE2蛋白过表达率及基因扩增率分别为16.67%(10/60)、23.33%(14/60)。HRE2蛋白过表达及基因扩增与胃癌发病年龄、组织学分型、浸润深度及淋巴结转移相关(均P<0.05)。结论 HER2可作为判断胃癌生物学行为和预后的参考指标。Objective To explore the relationship of protein expression and gene amplification of human epidermal growth factor receptor 2(HER2) with clinical pathological features of gastric cancer.Methods There were 60 patients with gastric cancer in this study.Protein expression and gene amplification of HER2 among both precancerous tissues and cancer tissues were examined using immunohistochemistry and fluorescence in situ hybridization(FISH).SPSS was used to conduct the analysis.Chi—Square test was used to compared between groups,and P<0.05 was considered as statistical significant.Results Negative protein expression and gene amplification of HER2 among precancerous tissues were not found in all 60 patients.The rates of HER2 over—expression and gene amplification among cancer tissues was 16.67%(10/60) and 23.33%(14/60),respectively.HER2 protein over—expression and gene amplification were related with age,histological type,depth of invasion,and lymph node metastasis(P<0.05).Conclusions HER2 may have close relationship with biological behavior and prognosis of gastric carcinoma.HER2 can be an important indicator for potential therapies among gastric cancer patients.
