摘  要:目的 探讨红藻氨酸 (KA)诱导性癫痫发作中相关脑区白细胞介素 6(IL 6)表达的时空规律和细胞学特征及其与c Fos的比较。方法 采用惊厥剂量KA(1 0mg·kg- 1 ,ip)诱导大鼠癫痫发作 ,记录行为 ,并分别取KA处理后 1 5、30min ,1、3、6、1 2及 2 4h的大鼠脑片 ,用免疫组化方法观察海马结构、梨状区及大脑皮层等相关脑区IL 6免疫反应 (IL 6ir)的变化特征 ,并与相应时空内的c Fos免疫反应 (Fos ir)相比较。结果 IL 6和c Fos均出现了在癫痫发作早期的快速表达 ,但IL 6ir的早期反应更为明显。KA后 1 5min海马结构内若干血管壁出现了明确的IL 6ir,30min时海马各亚区、梨状区、大脑皮层部位开始出现较强的IL 6ir阳性神经元 ,其数量明显多于阳性胶质细胞 ,KA后 3h以上各脑区IL 6ir进一步增强 ,6h后阳性神经元免疫反应性减弱 ,1 2~2 4h继续减弱 ,但阳性胶质细胞数量及IL 6ir强度变化不明显。与IL 6ir相比 ,Fos ir的变化较为短暂并且强度较弱 ,KA后 30min ,齿状回颗粒细胞层、海马各亚区锥体细胞层及梨状区等部位出现了微弱的Fos ir。结论 KA诱导癫痫发作过程中IL 6在相关脑区神经元内的表达迅速增加 ,明显早于c Fos。AIM To explore the expressional and cytological character of interlukin 6 (IL 6) in some brain regions using IL 6 immunoreactivity(IL 6 ir) and compare it with Fos immunoreactivity(Fos ir) in kainic acid (KA) induced seizure rats. METHODS The rats were given KA (10 mg·kg -1 , ip), and seizure behavior were observed. IL 6 and c Fos expressions were examined and compared using immunohistochmistry in hippocampus, piriform and cortex at 15 min, 30 min, 1, 3, 6, 12 and 24 h respectively after KA. RESULTS It was found that IL 6 ir occurred earlier and more intensively than Fos ir. IL 6 response to seizure was primarily located inside the blood vessels in some brain regions 15 min after KA; 30 min later, strong IL 6 ir positive neurons and a few glial cells were found in the layer of hippocampal pyramidal cells, piriform area and cortex. Stronger IL 6 ir and clear glial cells were still observable in the above brain areas up to 3 h following KA administration. At 6 h after KA, though IL 6 ir positive neurons became weak, glial cells were still clear. During 12 to 24 h, IL 6 ir still remained weak. Compared with IL 6 ir, Fos ir appeared later and remained shorter in time. Weaker Fos ir were found in granular cells of dentate gyrus and pyramidal cells in hippocampus, and piriform area at 30 min after KA. CONCLUSION IL 6 expression is dependent on time and severity of seizune induced by KA. IL 6 ir that appeares earlier and more intensive acts as an alarm signal and a marker of neuron damage caused by excitotoxin.
