摘  要:目的 :探讨脑缺血再灌损伤中雌激素与ET 3,GFAP的相互关系。 方法 :采用雌性昆明小鼠卵巢切除结合脑缺血再灌的动物模型及免疫组织化学方法。动物分为 3组 :①缺血再灌组 (IR) ,双侧颈总动脉结扎 7min ,再灌 1,3,5 ,14d ;②卵巢切除 /缺血再灌组 (OVX/IR) ,在缺血再灌前 1周作卵巢切除术 ;③假手术对照组。 结果 :对照组海马CA1区可见少量ET 3和GFAP免疫阳性细胞散在分布 ,IR组和OVX/IR组各时间点阳性细胞数明显增多 (P <0 .0 1)。缺血再灌 1d时 ,OVX/IR组ET 3和GFAP阳性细胞数明显高于IR组 (P <0 .0 1) ;5d时 ,OVX/IR组和IR组ET 3和GFAP阳性细胞数达高峰 ,在时空变化上存在一致性。 结论 :雌激素可能在缺血再灌的早期参与对ET 3和GFAP的调控而起保护作用。Objective: To detect the expression of ET 3 and GFAP immunoactivity in hippocampal CA1 of ovariectomied(OVX) mice following ischemia/reperfusion(IR), and to further explore the relationship of estrogen and ET 3 and GFAP.Methods: Fifty four adult female mice are randomly divided into three groups: ①IR group;②OVX/IR group. Ovariectomy was performed a week before bilateral common carotid arteries(BCCA) occlusion for 7 min. Each group above was subdivided into 1 d,3 d,5 d,14 d subgroups according to reperfusion time after the occlusion.③sham operated group, which was control group. Results: Few ET 3 and GFAP positive cells were diffusely distributed over the hippocampal CA1 area in control group, whereas the expression of ET 3 and GFAP was increased in both experimented groups. However the numbers of ET 3 and GFAP positive cells were significantly increased in OVX/IR group compared to IR group 1d after reperfusion( P< 0.01), the maximum increasing of ET 3 and GFAP was observed 5d after reperfusion. The increased expression of ET 3 was coupled with the activation of astrocyte.Conclusion: The withdrawal of estrogen caused by OVX might aggravate the expression of ET 3 and GFAP during early reperfusion. Therefore, estrogen probably plays a protective role in inhibiting astrocytic activation induced by ET 3.
