摘  要:目的:探讨丙酮酸(Pyruvate)对大鼠小肠缺血再灌注损伤的保护作用及其机制。方法:建立大鼠小肠缺血再灌注损伤的动物模型,实验组和对照组分别经肠腔给予含有丙酮酸钠和等能量的右旋糖酐-70营养液,对不同缺血再灌注时间的肠组织进行病理学观察并利用酶联免疫法检测肠组织中的肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)水平。结果:经丙酮酸处理后的小肠组织损伤情况:缺血45min为(2.17±1.17)分,再灌注30min为(2.17±0.98)分,再灌注60min为(2.33±1.03)分,较对照组明显减轻,P<0.01,t值分别为55.00,57.00,57.00。小肠黏膜组织中TNF-α和IL-6水平减低,TNF-α缺血45min,再灌注30,60min分别为(0.36±0.06),(0.87±0.06),(0.70±0.17)μg/L,与对照组比较P<0.01、F值分别为313.58,815.77,105.84。IL-6缺血45min,再灌注30,60min分别为(122.88±3.75),(213.37±8.91),(154.53±7.32)ng/L,与对照组比较P<0.01、F值分别为1587.18,1232.96,2447.93。结论:丙酮酸对大鼠小肠缺血再灌注损伤具有保护作用;该保护作用与丙酮酸减少小肠组织中的TNF-α和IL-6水平有关。AIM:To investigate the protective effect of pyruvate on ischemia-reperfusion injury in rat small bowel and its mech anism .METHODS:Rat model with ischemia-reperfusio n injury in small bowel was set u p.Instilled liquid diet containing pyruvate or isoenergetic polyglucose into th e lumen of the trial group or control group respectively.Histological studies o f intestine were carried ou t and tissue levels of tumor necrosis factorα(TNF -α)and interleukin 6(IL-6)were measured using enzyme-linked i m-munoso rbent assay at different time .RESULTS:The mean intestinal tissue injury grade s administered with pyruvate were(2.17±1.17),(2.17±0.98) ,(2.33±1.03)after 45min ischemia and 30min,60min reperfusion ,which were significantly slight er than the control group(t=55.00,57 .00,57.00respectively,P<0.01).The mean levels of TNF-αin admi nistered with pyruvate were(0.36±0.06),(0.87±0.06),(0 .70±0.17)μg /L after 45min ischemia and 30min,60min reperfusio n,which were s ignificantly lower than the control group(F=313.58,81 5.77,105.84respectively,P<0.01).The mean levels of IL-6were (122.88±3.75),(213.37±8.91),(154.53±7.32 )ng /L after 45min ischemia and 30min,60min reperfusion,which were sig nificantly lower than the contro l group(F=1587.18,1232.96,2 447.93respectively,P<0.01).Pyruvate has protective eff ect on is chemia-reperfusion in-jury in rat small bowel and the protective eff ect may be associated with de-creasing the levels of TNF-αand IL-6during th e intestinal tissu e injury.
