摘 要:目的 :探讨紫外线对体外培养的晶状体上皮细胞凋亡作用以及对晶状体水含量和超氧化物歧化酶 (superoxidedismutase,SOD)活性的影响 .方法 :紫外线诱导离体牛晶状体白内障形成 ,以TUNEL技术检测紫外线照射后培养 3,6 ,12 ,2 4h的晶状体上皮凋亡细胞 ,并测定晶状体水含量和SOD活性 .结果 :TUNEL法显示 ,照射组晶状体上皮细胞经紫外线照射后培养 6h出现凋亡细胞 ,对照组晶状体上皮细胞中无明显的凋亡细胞 ;照射组 12h后水含量明显升高 (P <0 .0 1)及SOD活性明显降低 (P <0 .0 1) .对照组水含量和SOD活性无明显变化 .结论 :紫外线诱导晶状体上皮细胞发生凋亡 ,并使晶状体水含量和SOD活性发生改变 。AIM: To study the effect of ultraviolet(UV) radiation on apoptosis of bovine lens epithelial cells in vitro and on lens water content and superoxide dismutase(SOD) activity. METHODS: Cultured bovine lenses were radiated by UV to establish cataract models in vitro . The apoptotic epithelial cells of lens were detected by TDT mediated digoxigenin dUTP nick end labeling (TUNEL) technique at 3, 6, 12 and 24 h after UV radiation respectively. At the same time, lens water content and SOD activity were measured. RESULTS: TUNEL staining showed that apoptotic cells appeared at 6, 12, 24 h after UV radiation. Few apoptotic cell was found in normal cells. Lens water content were increased ( P <0.01) and SOD activity were declined ( P <0.01) at 12 h after UV radiation. CONCLUSION: UV radiation can induce the apoptosis of lens epithelial cells and also affect lens water content and SOD activity. The apoptosis of lens epithelial cells induced by UV radiation is one of the early mechanisms in the development of cataract.
