摘 要:背景和目的:我们先前的研究提示,肝癌转移抑制基因可能在人染色体8p上。TEY1基因(TEnni-Yokusei1,转移抑制的日文)是最近从人染色体8p发现的前列腺癌转移抑制基因。本研究通过对不同转移潜能肝癌细胞株中TEY1基因的表达水平进行定量分析,探讨TEY1基因作为肝癌转移抑制基因的可能性。方法:提取肝癌细胞株MHCC97-H、MHCC97-L、HCCLM3和正常肝细胞系L02的RNA,反转录合成第一链cDNA,对反转录产物进行荧光定量PCR。用看家基因GAPDH的cDNA起始浓度作为参照标化TEY1的cDNA起始浓度,对标化结果进行单因素方差分析。结果:MHCC97-H、MHCC97-L、HCCLM3和L02标化后的TEY1cDNA起始浓度分别为(3.57±2.62)×10-4、(5.02±1.95)×10-4、(2.82±0.78)×10-5、(15.20±0.58)×10-4。肝癌细胞的TEY1基因表达水平和正常细胞之间存在显著性差异(P<0.001);HCCLM3的TEY1基因表达水平与MHCC97-H和MHCC97-L相比差异有显著性(P<0.05);MHCC97-L的TEY1基因表达水平是MHCC97-H的1.4倍,但两者无显著性差异(P>0.05)。结论:TEY1基因表达水平与肝癌细胞的转移潜能呈负相关,可以推测TEY1基因可能是肝癌转移的抑制基因。BACKGROUND &OBJECTIVE:The authorsprevious study sug g ested there could be metastasis suppressor g ene for liver cancer on h uman chromosome 8p.TEY1was a newly discovered metastasis suppressor g ene fo r prostate cancer located on human chr omosome 8p.In order to explore wheth er TEY1g ene could be a metastasis suppr essor g ene for liver cancer,the expression of TEY1was quantitated i n liver cancer cell strains with diff erent metastatic potential.METHODS :RNA was extracted from liver cancer c ell strains ,MHCC97-H,MHCC97-L,and HCCLM3,and normal liver cell line L02.The first strand cDNA was synthesize d by reverse transcription,which wa s then used as the template to perform fluor escent quantitative polymerase chain reaction(FQ-PCR).The quantity of the expression of TE Y1g ene was normalized by dividing with that of the housekeepi ng g ene,GAPDH for each sample.One-w ay analysis of variance(ANOVA)was performed for the normalized val ues.RESULTS :The normalized initial cDNA concent rations of TEY1in MHCC97-H,MHCC97-L,HCCLM3,and L02were(3.57±2.62)×10 -4 ,(5.02±1.95)×10 -4 ,(2.82±0.78)×10 -5 ,and(15.20±0.58)×10 -4 ,respectively.The expression level of TEY1in L02was sig nificantly different from those of liver cancer strains (P<0.001).The expression level of TEY1in HCCLM3was also sig nificantly different from those in MHCC97-H and MHCC97-L(P<0.05).But there was no sig nificant difference between MHC C97-H and MHCC97-L althoug h the expression level of TEY1g ene in MHCC97-L was 1.4times hig her than that in MHCC97-H.CONCLUSION:The expression levels of TEY1g ene in liver cancer cells were neg atively correlated wi th their different metastatic poten tials;it could be conjectured that TEY1may possibl y be a metastasis suppressor g ene for liver cancer.
