摘  要:目的 探讨无血清因素参与下血管内皮细胞 (Vasvularendothelialcell,EC)与细菌内毒素 (Lipopolysacchrides ,LPS)的结合特点。方法 用激光共聚焦显微镜观察异硫氰酸荧光素标记的内毒素 (FluoresceinisothiocyanatelabelledLPS ,FITC LPS)与EC的结合情况 ,并提取荧光标记内毒素与内皮细胞结合后细胞膜性成分观察其中的荧光物质强度。结果 EC与FITC LPS的结合存在浓度依赖性 ,结合的荧光主要集中于细胞浆 ,尤其是胞浆近核周区 ,核仁也可染色 ,结合的内毒素同细胞膜性成分关系密切。结论 EC可与LPS直接结合 ,并可能进入细胞内部而发挥作用。Objective To investigate the properties of the serum free binding of vascular endothelial cells(ECs) to lipopolysaccharide (LPS). Methods The binding of ECs to fluorescein isothiocyanate labelled LPS(FITC LPS) was observed with laser confocal microscope(LCM). The extract of ECs membrane after binding with FITC LPS was recorded with fluorescence microscope. Results The binding of ECs to FITC LPS was in a dose dependent manner. The fluorescence was mainly distributed in cytoplasm, especially near the nucleus which could also be stained. The bound FITC LPS had a very close relationship with the membranous system of ECs. Conclusion ECs could bind to LPS directly and could function intracellularly.
