摘  要:目的:探讨端粒酶及其亚单位(hTERT、TP1、hTR)在卵巢癌发生和发展中的作用,研究端粒酶活性与各亚单位表达的相关性。方法:应用逆转录-聚合酶链反应(reversetranscription-polymerasechainreaction,RT-PCR)技术检测66例卵巢上皮性肿瘤及10例正常卵巢组织端粒酶不同亚单位基因的表达情况,应用端粒重复序列扩增技术(telomeraserepeatamplificationprotocol,TRAP)检测其中45例卵巢上皮性肿瘤及9例正常卵巢组织端粒酶活性。结果:1)恶性、交界性卵巢肿瘤组织中端粒酶阳性检出率分别为83.33%(25/30)、71.43%(5/7),两组间无显著性差异(P>0.05),而良性上皮性卵巢肿瘤及正常卵巢上皮中未检测到端粒酶活性,与前两组之间均有显著差异(P<0.001);2)RT-PCR定性检测发现hTR和TP1mRNA在检测的各组组织中均有表达,而hTERTmRNA主要在上皮性卵巢癌和交界性卵巢肿瘤中表达,二者间hTERTmRNA表达的差别无统计学意义(P>0.05),而与良性及正常组织hTERT表达有显著差异(P<0.001);3)端粒酶活性与hTERT亚单位表达明显相关(P<0.001),而与hTR和TP1亚单位表达无明显相关性(P>0.05)。结论:端粒酶与卵巢癌发生发展过程密切相关;hTERT表达与端粒酶活性密切相关,并可能参与端粒酶的活性调节,hTERT的表达有望成为上皮性卵巢癌的诊断指标。Objective:This study is designed to investigate the role of telomerase and its subunits in the development of epithelial ovarian carcinoma,to ascertain the co-relationship be-tween telomerase activity and the expression of each subunit.Methods:Sixty-six epithelial ovari-an tumors and ten normal ovarian tissues were detected by reverse transcription-polymerase chain reaction(RT-PCR)for telomerase subunits,and telomerase repeat amplification protocol(TRAP)as-say was used to detect part of the samples,forty-five epithelial ovarian tumors and nine ovarian tissues for telomerase activity.Results:1)The positive rates of epithelial ovarian carcinomas and borderline ovarian tumors were83.33%and71.43%respectively,but none of the benign ovarian tumors and normal ovarian tissues were detected telomerase activity.The frequency of telomerase activity in ovarian carcinomas and borderline ovarian tumors was significantly higher than that in benign and normal tissues.2)It is found that hTR and TP1mRNA are widely expressed in each group of the tissues,while hTERT mRNA is mainly expressed in ovarian carcinomas and border-line ovarian tumors,and there is no significant difference between these two groups,which sug-gests that hTERT mRNA may play a critical role in the activation of telomerase.3)Statistics study showed that there is a strong correlation between the subunit hTERT and telom erase activity(P<0.001),whereas hTR and TP1expression showed no correlation with telomerase activation.Conclusion:Telomerase activation might contribute to the progression of ovarian carcinoma;be-cause hTERT mRNA expression parallels to telom erase activity,the hTERT may play a critical role in activation of telomerase;hTERT mRNA may be not only a useful diagnostic director but also a promising target for gene therapy in ovarian carcinoma .
