摘 要:目的 建立小鼠人乳腺癌自发性肺转移模型。方法 将人乳腺癌细胞株MDA MB 43 5s( 2× 10 5/只 )原位接种至 2 0只重症联合免疫缺陷 (SCID)小鼠 ,17周后处死 ,肉眼及显微镜下观察肺转移状况。结果 17周后存活的 8只小鼠中 4只 ( 5 0 % )肉眼下见肺转移灶 ,1~ 3个肺转移灶/只 ,直径 1~ 2mm ,显微镜下所有 8只小鼠均观察到肺转移。结论 将MDA MB 43 5s细胞原位接种至SCID小鼠可成功建立小鼠人乳腺癌自发性肺转移模型。Objective To establish a mouse model for human breast cancer spontaneous lung metastasis.Methods A human breast carcinoma cell line MDA-MB-435s was inoculated orthotopicly into exposed thoracic second mammary fat pads of 20 anesthetized SCID mice,2×105 cells per mouse.Mice were sacrificed 17 weeks after inoculation,and lung metastases were evaluated under both macroscopic and microscopic observations.Results Of the 8 mice survived within the experimental period,4 (50%) had macroscopic lung metastases,1-3 macrometastases per mouse,1~2 mm in diameter.Under microscopy,all 8 mice showed lung metastases.Conclusion A mouse model for human breast cancer spontaneous lung metastasis can be established by orthotopic inoculation of MDA-MB-435s cells into SCID mice.
