摘 要:目的 研究苦参碱对不同分化程度的胃癌细胞株在体外的杀伤作用及其机制。方法 用不同剂量的苦参碱分别作用于中、低分化胃癌细胞株SGC-7901和AGS 24、48、72h,以MTT法检测药物对肿瘤细胞的杀伤作用;透射电镜和流式细胞术检测苦参碱诱导SGC-7901凋亡的作用。结果 随药物浓度的增加和作用时间的延长,苦参碱对胃癌细胞的杀伤作用逐渐增强(P<0.01);低分化的AGS较中分化的SGC-7901更为敏感。透射电镜、流式细胞术检测均显示苦参碱能够诱导胃癌细胞凋亡。结论苦参碱在体外对胃癌细胞具有杀伤作用,且呈明显的量效和时效关系;低分化的胃癌细胞株AGS对苦参碱更为敏感;苦参碱对胃癌细胞株的体外杀伤作用与其诱导胃癌细胞凋亡有关。Objective To study the cytotoxicity effect of matrine and its mechanism on gastric cancer cell lines in different differentiations. Methods Matrine in different doses were used to treat moderate (SGC-7901) and lower (AGS) differentiated gastric cancer cell lines for 24、48 and 72 h respectively , and MTT assay was employed to evaluate the cytotoxicity of matrine on these tumor cell lines; meanwhile, transmission electron microscopy and flow cytometry were utilized to detect the apoptosis of SGC-7901 induced by matrine. Results Following the increase in the concentration of the drug and prolongation of administration times, the cytotoxicity of matrine on both kinds of gastric cancer cell lines were enhanced significantly(P<0. 01) ; especially the lower differentiated AGS was more sensitive to matrine than the moderate SGC-7901. Using transmission electron microscopy and flow cytometry, apoptosis induced by matrine was confirmed in SGC-7901. Conclusion The cytotoxicity of matrine on gastric cancer cell lines, in connection with its apoptosis-inducing effects, is dose and time dependent significantly; the lower differentiated gastric cancer cell line is more sensitive to matrine than the moderate one.
