摘 要:目的 观察染料木素和大豆苷元对去卵巢大鼠甘油三酯代谢的影响。方法 Wistar大鼠 70只 ,随机分成正常组、模型组、雌激素组、大剂量染料木素 (Genistein ,G)组、小剂量 (Genistein ,G)组、大剂量大豆苷元 (Daidzein ,D)组、小剂量 (Daidzein ,D)组 ,每组 10只。除正常组外 ,其余各组均切除双侧卵巢。术后 1wk开始给药染料木素和大豆苷元 ,6wk后处死动物 ,测定血清甘油三酯 (sTG)浓度、肝脏组织甘油三酯 (hTG)含量、肝脏组织磷脂含量和磷脂指数 ,并固定肝脏组织、HE染色 ,进行病理学观察。结果 大鼠切除卵巢6wk后hTG含量明显增加 ,HE染色发现肝脏细胞中分布大小不一的脂滴 ;肝脏磷脂指数降低。雌激素和大豆苷元均可逆转以上变化 ,但雌激素升高sTG浓度并且对肝细胞造成空泡样变损伤 ;染料木素对各项指标均无明显作用。结论 大豆异黄酮中影响去卵巢大鼠甘油三酯代谢的有效成分为大豆苷元 ,其可以有效抑制去卵巢大鼠肝脏组织中脂肪的堆积。AIM To study effects of genistein and daidzein on triglyceride metabolism of ovariectomized rats. METHODS 70 Wistar rats were randomly divided into 7 groups: control group, model group, estrogen group, genistein group of high dose and low dose, daidzein group of high dose and low dose,with 10 rats in each group. All rats were ovariectomized except that of the control group. One week after operation , the soy isoflavones were administrated with different doses of genestein and daidzein for 6 weeks. Seven weeks after operation, the rats were killed, with serum and liver taken, and the levels of serum triglyceride (sTG), liver homogenate triglyceride (hTG) and phospholipids, and phospholipids index were measured; then a part of liver organ was fixed,and its pathological changes were observed under microscope. RESULTS The content of sTG in liver of ovariectomized rats of model group increased significantly, there were lots of lipid droplets in its liver cells seven weeks after operation. At the same time, the phospholipid index of ovariectomized rats decreased. Administration of estrogen ordaidzein could attenuate the phospholipial index in ovariectomized rats,but estrogen increased the level of sTG and had harm to liver cells. Genistein did not have any effects. CONCLUSION The effective component of soy isoflavone on triglyceride metabolism of ovariectomized rats is daidzein, which suppresses the increase of triglyceride content in liver induced by ovariectomizing.
