摘  要:目的 探讨VEGF、VEGFmRNA在局灶脑缺血中心区和边缘区的表达和动态变化。方法 SD大鼠54只。根据缺血时间和再灌时间分为 9组 ,采用线栓并环扎的方法建立局灶脑缺血再灌模型。LSAB法免疫组化染色观察VEGF免疫阳性表达。RT PCR法检测VEGFmRNA动态变化。结果 在缺血 3~ 6h ,缺血中心区与边缘区VEGF免疫阳性反应达高峰 ,VEGFmRNA转录水平明显升高 ,两者 2 4h基本降至正常水平。Objective Dynamic observation the expressions and changes of VEGF and VEGF mRNA in marginal and central areas of ischemic region at different times and studying the relationships between them. Methods 54 SD rats were divided into 9 groups according to ischemic and reperfusion time. The ischemic model was established by the method of embolizing and fastening cerebral vessels. VEGF was observed by the method of immunohistochemical stain. Dynamic changes of VEGF mRNA were observed using the method of RT PCR. Results After 3~6hrs' ischemia,the positive reactions of VEGF reached peak in ischemic centres and marginal areas and transformation levels were elevated obviously. Those changes reduced to normal level in 24hrs. Conclusion The high expressions of VEGF in the early stage of ischemia maybe have a close relationship with the self protection of neurons and endotheliocytes.
