摘  要:目的 探讨部分去背根后备用背根节 (L6 )各类细胞脑源性神经营养因子 (BDNF)及其mRNA的量变情况。 方法 对成年雄性猫行单侧部分背根切断术 (切除一侧L1 ~L5、L7~S2 DRG ,保留L6 为备用根 )。取正常组一侧和术后 3d及 7d组手术侧的L6 DRG并分别行免疫组织化学及原位杂交染色。观察BDNF及其mRNA在DRG各类细胞的分布 ,测定BDNF及其mRNA在神经元和卫星细胞的光密度值。 结果 部分去背根后 3d ,中小神经元内BDNF及其mRNA的光密度值较正常者明显减少 (P <0 0 5 ) ,而 7d时又恢复近正常水平 (P >0 0 5 )。BDNF及其mRNA在大神经元的光密度值术后 3d无明显变化 ,而 7d时较正常者明显减少 (P <0 0 5 )。比较之 ,卫星细胞BDNF及其mRNA的光密度值术后 7d时明显升高 (P <0 0 5 ) ,术后 3d与正常者比未见明显变化。 结论 部分背根切断导致备用DRG各类细胞BDNF的表达水平发生不同程度的改变。提示备用DRG内BDNF的变化与脊髓Ⅱ板层可塑性有关。Objective To explore the change of BDNF and its mRNA in spared root ganglion following partial dorsal rhizotomy. Methods Fifteen male adult cats were divided into three groups.Two group animals were performed unilateral partial dorsal root rhizotomy(L 1-L 5,L 7-S 2 DRG were sectioned,but L 6 was spared) and animals were sacrificed at survived 3 days and 7 days after operation.L 6 dorsal root ganglion(DRG) of operation side and control from each animal was taken and sectioned into 20 μm thickness.The sections were stained by immunohistochemistry method(BDNF antibody 1∶500,Santa cruz) and in situ hybridization technique(BDNF cRNA probe 1∶100).The A value of BDNF and its mRNA in the large,medium and small sized neurons as well as satellite cells in DRG in one side of normal animals,operation side of unilateral dorsal rhizotomy was counted and analyzed by statistic method(q test). Results The positive product of BDNF and its mRNA in L-6DRG was mainly observed in medium and small sized neurons(13-57?μm) and a few large sized neurons(>57?μm).In addition,the positive product of BDNF was also observed in a few satellite cells.At 3 days after partial dorsal rhizotomy,the A value of BDNF and its mRNA in the medium and small sized neurons decreased apparently than that in normal group(P<0 05),while recovered to normal level at 7 days after operation(P>0 05).On the contrary,the A value of BDNF and its mRNA in the large sized neurons has not apparent changed at 3 days after partial dorsal rhizotomy but decreased apparently at 7 days after operation(P<0 05).In contrast,the A value of BDNF and its mRNA in the satellite cells increased apparently at 7 days after operation(P<0 05).However,apparently changes in the satellite cells were not seen at 3 days after operation.Conclusion Our results suggest that the BDNF expression among three type of neurons and satellite cells in DRG following partial dorsal root rhizotomy was changed.The results suggest that the role of BDNF in DRG may involve in the plasticity of
