摘 要:目的 :探讨STI5 71对ANLL白血病细胞c-kitmRNA表达及细胞分化的影响。方法 :不同浓度STI5 71培养前后逆转录 -聚合酶链反应方法测定c -kitmRNA的表达 ,涂片吉姆萨染色及过氧化物酶染色观察细胞的分化情况。结果 :①培养后不同药物浓度组的粒细胞及单核细胞均有不同程度的分化 ,随着药物浓度的增加 ,原早幼粒细胞逐渐减少 ,中晚幼粒和成熟单核细胞数增加 ,有诱导向同系成熟分化作用 ,并且过氧化物酶染色阳性程度增加 ;②c -kit受体mRNA的表达培养前 2 .2± 0 .4 9,培养后 (对照、0 .1、1、10 μmol·L-1)浓度组c-kitmRNA表达的平均值分别为 2 .1± 0 .6、1.2± 0 .4、0 .8± 0 .4、0 .5± 0 .2 ,培养前与培养后各药物浓度组有显著性差异 (P <0 .0 5 ) ,各浓度组与对照组之间差异显著 (P <0 .0 5 ) ,且 0 .1μmol·L-1与 10 μmol·L-1两个浓度组之间差异显著 (P <0 .0 5 )。结论 :STI5 71对急性非淋巴细胞白血病原代细胞有诱导向同系成熟分化作用 ,对c -kit酪氨酸激酶有明显的抑制作用 ,且与药物浓度相关。Objective: To explore the effect of STI571 on the expression of c-kit and cell differentiation of the bone marrow mononuclear cells from patients with acute non-lymphocytic leukemia(ANLL).Methods:The cells were exposed to various concentration of STI571 for 72 hours in vitro,and the expression of c-kit mRNA was assayed by RT-PCR,the cell differentiation were studied by Giemsa staining and peroxidase staining. Results:①After treated by different concentration of STI571 , there were an iso-degree differentiation in granulocytic cell and monocyte. With the increased concentration of STI571 , progranulocyte and promyelocyte gradually decreased, myelocyte , metamyelocyte , mature monocyt and the positive peroxidase staining increased. ②Before STI571 cultivation,the mRNA expression of c-kit re ceptor was 2.2±0.49.After cultivation, the expressions of different concentrations (control, 0.1 μmol·L -1, 1 μmol·L -1, 10 μmol·L -1) were 2.1±0.6,1.2±0.4,0.8±0.4,0.5±0.2 respectively.It was significantly lower than that of the group before treatment and controls (P<0.05), the expression of 0.1 μmol·L -1 STI571 group was significantly higher than that of 10 μmol·L -1group(P<0.05). Conclusion: STI571 inhibited c-kit and induced cells toward iso-series mature differentiation, which relies on the concentration of STI571 .
