摘 要:目的 :观察灯盏花素注射液对沙土鼠脑缺血再灌注后能量代谢和脑水肿的影响及其防治作用机制。方法 :制备沙土鼠前脑缺血再灌注模型 ,脑缺血时间为 10 min。将实验动物随机分成假手术组、常温对照组和灯盏花素组 ,根据再灌注时间将后两组又分 4个亚组 ,即缺血末组、再灌注 10 min组、再灌注 30 m in组和再灌注 6 0 m in组 ,每个亚组 8只动物。脑水的测定采用干湿重法 ,应用高效液相及紫外检测仪测定海马 ATP、ADP、AMP的含量。缺血再灌注期间将脑温始终控制在 (37.0± 0 .2 )℃。结果 :常温对照组各再灌注时间点海马 ATP含量和腺苷酸池 (ATP+ADP+AMP)含量均明显低于假手术组 (P均 <0 .0 5 ) ,而灯盏花素组海马ATP和腺苷酸池 (ATP+ADP+AMP)含量均明显高于常温组 (P均 <0 .0 5 )。在常温对照组脑水的含量明显大于假手术组 (P<0 .0 5 ) ,而灯盏花素组脑水的含量尽管高于假手术组 ,但明显低于常温组 (P<0 .0 5 )。结论 :灯盏花素注射液可能通过抑制能量代谢障碍、减轻脑水肿而发挥脑保护作用。Objective: To study the effect of breviscapine on energy metabolism and cerebral edema after cerebral ischemia/reperfusion in gerbils and to explore the mechanism of breviscapine attenuating cerebral ischemia/reperfusion injury.Methods:Forebrain ischemia in gerbils was produced by occlusion bilateral common carotid arteries with aneurysm clamps for 10 minutes. Gerbils were randomly divided into sham- operated group, normothermia group and breviscapine group. Both normothermia group and breviscapine group were further divided into four subgroups according to the reperfusion time (0, 10, 30 and 60 minutes). The contents of ATP, ADP and AMP were determined by high liquid chromatography (n=8). Brain water was determined by drying electrothermostat. The brain temperature in all gerbils during cerebral ischemia/reperfusion was kept at (37.0±0.2)℃. Results: ATP and adenine nucleotide pool levels in hippocampus in normothermia group significantly decreased after reperfusion (all P<0.05). ATP and adenine nucleotide pool levels in hippocampus in breviscapine group were much more than those in normothermia group after reperfusion (all P<0.05). The content of brain water in normothermia group significantly increased after reperfusion (P<0.05). However the content of brain water in breviscapine group was much less than that in normothermia group after reperfusion (P<0.05). Conclusion: Breviscapine could attenuate the failure of energy metabolism, attenuate cerebral edema during cerebral ischemia/reperfusion, which may be the results of its neuroprotection during cerebral ischemia/reperfusion.
