胃癌组织端粒酶逆转录酶和Rb基因表达及其临床意义

摘　　要：目的 :探讨端粒酶逆转录酶 (humantelomerasereversetranscriptase ,hTERT)、Rb蛋白在胃癌中表达。方法 :应用SP法检测了 5 0例胃癌标本 ,2 5例胃癌前病变组织 ,11例正常胃黏膜组织。结果 :胃癌组hTERT阳性率为 86 0 %。胃癌组Rb表达阳性率为 5 8 0 %。hTERT活性表达与胃癌浸润深度、淋巴结转移及临床分期呈显著正相关 ,P <0 0 5。Rb表达与胃癌淋巴结转移、临床分期呈显著负相关 ,P<0 0 5。hTERT表达与Rb表达呈显著负相关 ,P <0 0 5。结论 :hTERT表达与胃癌的发生高度相关 ,检测hTERT有助于胃癌的早期诊断 ;Objective To investigate the role of hTERT and Rb protein expression in gastric cancer.Methods The expression of hTERT and Rb protein was measured in formalin-fixed,paraffin-embedded gastric cancer specimens from 50 patients in gastric cancer group,25 patients in precancerous lesions group,and 11 healthy people in normal control group by using the strept avidin biotin peroxidase method.Results The positive rate of hTERT protein expression in gastric cancer group was 86.0%,Rb protein were expressed in 58.0% of gastric cancer group,The positive expression of hTERT was correlated with the degree of depth of invasion,lymph node metastasis and clinical stage of gastric carcinoma,P<0.05.The abnormal expressions of Rb were correlated with the lymph node metastasis and clinical stage of gastric carcinoma,P<0.05.Conclusions There are a perfect positive correlation between the expression of hTERT and carcinogenesis of gastric cancer and a perfect negative correlation between the expression of Rb protein and carcinogenesis of gastric cancer.The detection of hTERT may be helpful in early diagnosis of gastric cancers.The detection of hTERT and Rb protein may also be helpful in evaluating the prognosis of gastric cancers.The expression of hTERT is negatively correlated with the expression of Rb,P<0.05.It suggests that the deficiency of Rb protein be one of the acting pathways of hTERT.
