摘  要:目的 探讨环氧合酶 (COX) 2反义寡核苷酸 (ODNs)对胰腺癌PC 3细胞株COX 2mRNA和蛋白表达的影响 ,并进一步阐明COX 2反义ODNs基因治疗胰腺癌的可行性。方法 设计和合成COX 2反义ODNs,体外转染胰腺癌PC 3细胞 ,然后通过荧光显微镜、RT PCR及Westernblotting证实转染效果。结果 转染COX 2反义ODNs后 ,PC 3细胞COX 2mRNA和蛋白表达均下降 ,且体现出一定的剂量和时间依赖性关系。结论 COX 2反义ODNs转染胰腺癌PC 3细胞株后 ,可下调细胞中COX 2mRNA和蛋白表达 ,COX 2反义ODNs有可能达到胰腺癌基因治疗的效果。Objective To investigate the effects of COX-2 anti-ODNs on the expression of COX-2 mRNA and protein in pancreatic carcinoma PC-3 cell line, and to clarify the possibility of COX-2 anti-ODNs gene therapy on pancreatic carcinoma. Methods The COX-2 anti-ODNs was designed, synthesized, and transfected into PC-3 cell line.The transfection effects were confirmed by fluorescence microscopy, RT-PCR and Western blotting. Results The expression of COX-2 mRNA and protein was decreased after transfecting COX-2 anti-ODNs in PC-3 cell line in a certain degree of time-dependent and dose-dependent manner Conclusion The expression of COX-2 mRNA and protein in PC-3 cell line can be down-regulated by COX-2 anti-ODNs, which suggests COX-2 anti-ODNs can exert the effects of gene therapy on pancreatic carcinoma.
