摘  要:目的观察移植骨髓间充质干细胞(BM-MSCs)在大鼠瘢痕子宫模型内的分布情况,建立可供动态观察的瘢痕子宫模型。为今后进一步研究改善瘢痕子宫功能的方法提供一定实验基础,以及了解BM-MSCs对瘢痕子宫的修复作用。方法利用梯度密度离心结合贴壁培养法分离和培养BM-MSCs,传代至第4代用流式细胞技术对其进行检测,之后利用腺病毒-绿色荧光蛋白标记干细胞。建立大鼠瘢痕子宫模型,经尾静脉注射经标记的干细胞悬液,观察干细胞在子宫切口部位的分布以及愈合情况。结果子宫切口部位可见移植后的干细胞聚集,并能减轻瘢痕愈合。结论腺病毒—绿色荧光蛋白标记技术可以示踪干细胞在活体内的分布,移植干细胞在一定程度上能减轻子宫的瘢痕愈合,促进修复。Objective To observe the distribution of bone marrow mesenchymal stem cells( BM-MSCs) in rat model of scarred uterus,establish a scarred uterus model for dynamic observation,provide a certain experimental basis for further researching the methods to improve the function of scarred uterus,investigate the recovery effect of BM-MSCs for scarred uterus. Methods BM-MSCs were isolated and cultured with gradient density centrifugation and adherent culture,then BM-MSCs of the fourth generation were detected by flow cytometry.Adenovirus-green fluorescent protein was used to label stem cells. Rat model of scarred uterus was established,the labeled stem cell suspension was injected through the tail vein. The distribution of stem cells and healing in incision site of the uterus were observed. Results Stem cell aggregation after transplantation can be observed in uterine incision,and scar healing was promoted. Conclusion Adenovirusgreen fluorescent protein labeling technique can trace the distribution of stem cells in the living body. The transplanted stem cells can heal scar of the uterus to a certain extent and promote repair.
