摘  要:目的:观察miR-145表达对胶质瘤U251干细胞侵袭力的影响并探讨其可能机制。方法:体外培养从U251细胞中分离的CD133+细胞,分别转染miR-145拟似物和miR-145抑制物,实时定量PCR法检测转染后U251干细胞中miR-145的表达水平。转染24 h后,Transwell试验评价U251干细胞侵袭能力的变化;转染48 h后,蛋白质印迹法检测侵袭相关基质金属蛋白酶(matrix metalloproteinase,MMP)-2、MMP-9和金属蛋白酶组织抑制因子-3(tissue inhibitor of metalloproteinase-3,TIMP-3)蛋白的表达。结果:转染miR-145拟似物能有效提高U251干细胞中miR-145的表达,抑制U251干细胞侵袭能力,下调其MMP-2、MMP-9及上调TIMP-3的表达(P<0.05);转染miR-145抑制物能有效降低U251干细胞中miR-145的表达,增强U251干细胞侵袭能力,上调其MMP-2、MMP-9的表达并下调TIMP-3的表达(P<0.05)。结论:miR-145表达水平与胶质瘤干细胞的侵袭能力关系密切。Objective:To investigate the function and possible mechanisms of miR-145 on the invasion of glioma U251 stem cells.Methods:CD133 + cells isolated from the U251 cells were cultured in vitro and transfected with miR-145 mimics and miR-145 inhibitor.The expression of miR-145 was detected by real-time PCR.After 24 h transfection,the invasive effects of U251 stem cells were measured by Transwell assay, and the expressing levels of matrix metalloproteinase(MMP)-2,MMP-9 and tissue inhibitor of metalloprotei-nase-3(TIMP-3)were examined by Western blotting after 48 h transfection.Results:miR-145 mimics could effectively improve the expression of miR-145 in U251 stem cells,inhibit the invasion of U251 stem cells,and downregulate MMP-2 and MMP-9 protein expression and upregulate TIMP-3 protein expression(P<0.05);while miR-145 inhibitor could effectively decline the expression of miR-145 in U251 stem cells, enhance the invasion of U251 stem cells,and upregulate MMP-2 and MMP-9 protein expression and down-regulate TIMP-3 protein expression(P <0.05).Conclusion:The expression of miR-145 is related with the invasion of glioma stem cells.
