摘 要:目的观察绞股蓝皂苷(GPS)对2型糖尿病(T2DM)并非酒精性脂肪性肝病(NAFLD)大鼠肝组织脂质沉积量的影响。方法 65只SPF级雄性SD大鼠,体重220~250 g,随机分为空白对照组(7只,N组),NAFLD模型组(NM组,7只),T2DM并NAFLD模型组(51只)。N组给予普通饲料喂养。NM组给予高脂饲料喂养。模型组先给予高糖高脂饲料喂养4 w;隔夜空腹腹腔注射链脲佐菌素(STZ)40 mg/kg,继续给予高糖高脂饲料喂养至8 w,建立T2DM并NAFLD大鼠模型;将模型组分为GPS大剂量干预组(9只,JH组),予GPS 1 g·kg-1·d-1灌胃;GPS小剂量干预组(9只,JL组),予GPS 0.5 g·kg-1·d-1灌胃;T2DM并NAFLD模型组(9只,M组),同等体积纯净水灌胃;继续给予高糖高脂饲料;治疗周期6 w。实验周期14 w。检测血甘油三酯(TG)、胆固醇(TC)、血糖(GLU)、肝组织TG、肝脏病理学。结果肝组织TG水平:JH组、JL组低于M组,比较有显著差异(P<0.01)。血糖水平NM组与N组存在显著差异(P<0.01)。血TG水平:JH组、JL组与M组有显著差异(P<0.01)。肝脏病理学:NM组、M组重度NAFLD,JH组、JL组中至重度NAFLD。结论绞股蓝能够降低T2DM并NAFLD大鼠肝脏脂质沉积量。Objective To investigate the effect of Gypenosides on rat liver tissue lipid deposition with type 2 diabetes Mellitus (T2DM) and nonalcohol fatty liver disease (NAFLD) and the mechanisms.Methods 65 SPF male SD rats, body mass 220~250 g, were randomly divided into blank control group (group N, n=7), NAFLD model group (group NM, n=7), NAFLD and T2DM model group (n=51).Rats in NAFLD and T2DM model group were divided into three groups: high dose GPS group (JH, n=9) treated with 1 g· kg-1· d-1 GPS, low dose GPS group (JL, n=9) treated with 0.5 g· kg-1· d-1 GPS, model group (M, n=9) treated with the same volume of water , and high fat diet at the same time.Plasma triglycerides ( TG) and total cholesterol ( TC) , blood sugar ( BS) , TG in liver tissue and liver pathology were detected.Results TG level in group JH and group JL were lower than that of group M ( P<0.01 ).TG level in group JH was lower than that of group JL , the difference was significant ( P<0.05 ).BS level of group JH and group JL were lower than that of group M ( P<0.01 ).BS level of group JH was lower than that of group JL ( P<0.05 ).The plasma TG level of group JH and group JL were lower than that of group M ( P<0.01 ).The plasma TG level of group JH was lower than that of group JL , the difference was significant ( P<0.05 ).Liver pathology of NM group , M group developed severe fatty liver and became moderate to severe degree of NAFLD in group JH, group JL.Conclusions Gypenosides could reduce lipid deposition in the liver in rat with T 2DM and NAFLD.
