摘 要:目的 :研究光动力抑癌作用机制。方法 :采用3H -TdR掺入法检测激光光敏化BPD -MA后人膀胱癌细胞株BIU -87细胞DNA的合成。结果 :实验前对照组的CPM值为 482 3 .5± 5 4.2 6,激光激活BPD -MA光动力显著抑制人膀胱癌细胞株BIU -87细胞DNA的合成 (12 63 .7± 2 0 6.84VS 482 3 .5± 5 4.2 6,P <0 .0 5 )。而单纯光敏剂BPD -MA和假照射处理对人膀胱癌细胞株BIU -87细胞DNA合成无明显影响 (P >0 .0 5 )。结论 :激光激活BPD -MA光动力抑制人膀胱癌细胞株BIU -87细胞DNA的合成可能是其抑癌作用机制之一。Objectives:To study on the mechanisms of photodynamic therapy on malignant tumors.Method:Photosensitization of BPD-MA was activated by laser with red light(632.8nm)delivered at 10 mW/cm 2 to give a total dose of 2.4J/cm 2.DNA synthesis of human bladder cancer BIU-87 cells was determined with 3 H-TdR incorporation.Results:A baseline proliferative activity in BIU-87 cells before laser irradiation without BPD-MA was maintained at a rate of 3 H-thymidine incorporation of 4823.5±54.26CPM.Treatment of cells with photodynamic therapy significantly decreased cellular proliferative activity as measured by 3 H-thymidine incorporation (1263.7±206.84 VS 4823.5±54.26, P<0.05).However,Laser irradiation alone,BPD-MA alone and sham radiation without BPD-MA did not affect the DNA synthesis of human bladder cancer BIU-87 cells (P>0.05).Conclusion:Treatment of cells with photodynamic therapy significantly decreases cellular proliferative activity as measured by 3 H-thymidine incorporation,which is one of the mechanisms of photodynamic therapy on malignant tumors.
