摘 要:【目的】研究喉鳞癌组织中p73,p5 1基因的表达情况及其临床意义。【方法】应用反转录聚合酶链反应技术 (RT PCR)检测 4 3例喉鳞癌组织 ,17例喉鳞癌旁组织及 2 4例声带息肉组织标本中 p73和p5 1A ,p5 1BmRNA的表达情况。【结果】4 3例喉鳞癌组织中 2 6例p73mRNA表达阳性 ,17例对应喉癌旁组织中 4例 p73mRNA表达阳性 ,2 4例声带息肉组织标本中 8例 p73mRNA表达阳性。喉鳞癌组织中p73mRNA的表达与喉癌旁组织、声带息肉组织相比 ,其差异具有显著性 (P <0 .0 5 )。喉鳞癌组织中 p73mRNA的表达与TNM分期及复发存在密切关系 ,Ⅲ~Ⅳ期及复发组患者中 p73mRNA的表达较Ⅰ~Ⅱ期及初发组患者的表达强 (P <0 .0 5 )。p5 1A ,p5 1BmRNA在所检测的标本中均有表达。其中 ,p5 1A、p5 1BmRNA在喉癌组织中的表达较癌旁组织及声带息肉组织中的表达强 (P <0 .0 5 )。喉鳞癌组织中 ,p5 1AmRNA的表达较 p5 1BmRNA的表达强 (P <0 .0 5 )。p5 1A ,p5 1BmRNA的表达与所有的临床病理参数均无关。【结论】喉鳞癌组织中存在 p73,p5 1基因的过度表达现象 ,p73,p5 1基因的异常表达与喉鳞癌的发生发展存在一定的关系。p5 1基因可作为鳞状上皮源性肿瘤的重要标记物。[Objectives]To investigate the expression of p73 and p51 genes in laryngeal squamous cell carcinoma (LSCC) and its clinical significance.The expressions of p73 mRNA, p51A / p51B mRNA in 43 human LSCC tissues,17 para-cancerous tissues and 24 polyp specimens from vocal cord tissues were detected by reverse transcriptase polymerase chain reaction (RT-PCR) techniques.Overexpression of p73 mRNA were found in 26 out of 43 LSCC tissues, in 4 out of 17 para-cancerous tissues and in 8 out of 24 polyp specimens from vocal cord tissues. The positive expression rate of p73 mRNA in LSCC tissues was significantly higher than that in para-cancerous tissues and polyp tissues of vocal cord (P<0.05).The positive expression rate of p73 mRNA in LSCC tissues was closely related to the stages of tumor, nodes and metastasis (TNM) system classification and recurrence. The expression of p73 mRNA in group of patients with stages Ⅲ~Ⅳ and recurrence was significantly higher than that in patients with stages Ⅰ~Ⅱ and earlier involvement (P< 0.05 ). Expressions of p51A mRNA and p51B mRNA were found in all detected tissues. Much stronger expression of these mRNAs existed in LSCC tissues as compared with that in para-cancerous tissues and polyp specimens of vocal cord (P<0.05). The expression of p51A mRNA in LSCC tissues was much stronger than that of p51B mRNA (P<0.05). There was no relationship between the expression of p51A/p51B mRNA and all clinico-pathological parameters.[Conclusion]Overexpression of p73 and p51 mRNAs occurs in LSCC tissues and certain relationship is observed between their abnormal expression and oncogenesis /development of LSCC. p51 gene may be an important marker of tumors from squamous epithelium origin.
