摘 要:目的建立蓝布正中总鞣质、没食子酸及鞣花酸含量的测定方法。方法以干酪素作为吸附剂,磷钼钨酸为氧化剂,没食子酸为对照品,在碱性条件下显色,建立紫外可见分光光度法测定蓝布正中总鞣质含量的方法;采用高效液相色谱(HPLC)法同时测定蓝布正中没食子酸和鞣花酸的含量,使用Agilent Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),流动相为0.01%甲酸水溶液-甲醇,梯度洗脱,流速1.2 mL·min-1,柱温30℃,检测波长274 nm。结果以没食子酸计,总鞣质含量在1.018 0~10.180 0μg·mL-1范围内线性关系良好,平均回收率为99.82%,相对标准偏差(RSD)为1.75%。没食子酸和鞣花酸分别在2.405 0~54.113 0、5.275 0~118.690 0μg·mL-1呈良好线性关系,平均回收率为99.6%、99.3%,RSD分别为1.14%、0.88%。结论本研究所建方法操作简便、准确、重复性好,可更全面地反映蓝布正药材中的鞣质特征。Objective To establish a method for determining the contents of total tannins,gallic acid and ellagic acid in Geum Japonicum Thunb.var.Chinese F bolle.(Chinese name Lanbuzheng,LBZ).Methods The UV-vis spectrophotometry was used to determine the content of total tannins of LBZ with casein as sorbent,phosphorus molybdenum-tungsten acid as oxidant.Contents of gallic acid and ellagic acid were determined by HPLC.The separation was performed on the Eclipse XDB-C18 column(250 mm×4.6 mm,5μm)with gradient elution.The mobile phase was 0.01%formic acid water solution and methanol,and the flow rate was set as 1.2 mL·min-1.The column temperature was kept at 30℃.The detection wavelength was set at 274 nm.Results For determination of total tannins,the linear relationship of standard curve was good within the range of 1.018 0-10.180 0μg·mL-1,and the average recovery was 99.82%with RSD of 1.75%.The linear ranges of gallic acid and ellagic acid were within2.405 0-54.113 0,5.275 0-118.690 0μg·mL-1,respectively.The average recovery rates for gallic acid and ellagic acid were 99.6%and 99.3%with RSD of 1.14%and 0.88%.Conclusion These methods are simple,accurate and suitable for the determination of the total tannins,gallic acid and ellagic acid in LBZ.
