摘 要:用32P放射性贴片照射HeLa细胞,细胞化学染色和生长曲线校正相结合检测照射后表达衰老相关性β-半乳糖苷酶(SA-β-Gal)的细胞及其数量变化趋势.结果表明,照射后,部分肿瘤细胞可以逆转进入表达SA-β-Gal的死亡相M1或M2.由照射诱发的染色阳性细胞的绝对数量在观察时间内持续增长,并且,在一定剂量率范围内(7.5-60cGy/h)的等剂量照射后,低剂量率β射线可以诱发更多的SA-β-Gal阳性细胞.此外,诱发衰老是核素内照射治疗肿瘤中存在的远期治疗效应的可能机制.HeLa cells were irradiated with 32 P radioactive filter papers. Cells expressing senescence associated β-galactosidase(SA-β-Gal) were quantified. And to acquire information about the absolute numbers of SA-β-Gal+ cells and their changing tendency, the rates of cells expressing SA-β-Gal were corrected with cell growth curves. Results show that after low-dose-rate β irradiation, some tumor cells can reverse into mortality stage M1 or M2 cells which express SA-β-Gal. Numbers of SA-β-Gal+ cells induced by the irradiation were increased. And in dose rates of 7.5—60cGy/h, more SA-β-Gal+ cells could be induced by the irradiation than higher dose-rate irradiations at the same dose. Moreover, it may indicate that long-term therapeutic effect of internal irradiations could be resulted by induced aging of tumor cells .
