β-连环蛋白和CyclinD1与胰腺癌的生物学意义

摘　　要：目的　研究 β cat与CyclinD1蛋白的表达与胰腺癌发生和生物学行为的关系。 方法　应用免疫组织化学SABC法检测 42例胰腺癌组织和 2 0例正常胰腺中 β cat和CyclinD1的表达。 结果　β cat在 2 0例正常胰腺均呈正常表达 ,在 42例胰腺癌组织中异常表达率为 69 0 5 % ( 2 9/ 42 ) ,胰腺癌的异常表达率显著高于正常胰腺 (P <0 0 1) ,而且与病人年龄 ,性别 ,肿瘤大小 ,分化程度无关 (P >0 0 5 )。与临床分期和淋巴结转移显著相关 (P <0 0 1)。CyclinD1在 42例胰腺癌过表达率为 71 43 % ( 3 0 / 42 ) ,明显高于正常胰腺 (P<0 0 1)。CyclinD1的过表达与病人年龄 ,性别 ,肿瘤大小 ,病理分级 ,临床分期 ,淋巴结转移均无关 (P >0 0 5 )。β cat的异常表达与CyclinD1的过表达显著相关 (P <0 0 5 )。结论　β cat参与胰腺癌的发生 ,并有可能通过激活CyclinD1使细胞增殖 ,促使肿瘤发生。Objective To investigate the abnormal expression of β-catenin and CyclinD1 in pancreatic carcinomas and its relationship with the biological behavior.Methods Immunohistochemistry (SABC method ) was used to detect β-catenin and CyclinD1 in 42 pancreatic carcinomas and 20 normal pancreatic tissues.Results β-catenin normal expression was detected in 20 normal pancreatic tissues.The abnormal rate of β-catenin expression in pancreatic carcinomas was 66.7%(28/42).The significant relationship was observed between β-catenin expression and clinical stages and lymph node spread (P<0.01).Compared with the normal pancreatic tissues,the overexpression of CyclinD1 in pancreatic carcinomas was 69.05%(29/42). There was a significant difference(P<0.01).No significant difference was found between the abnormal expression of β-catenin and CyclinD1 and the patients'age, sex,tumor size,and differentiation (P>0.05).There was a significant relationship between the abnormal expression of β-catenin and CyclinD1.Conclusions β-catenin plays an important role in the tumorigenes of pancreatic cancer and may improve the cells proliferation by transactive cyclinD1.
