摘 要:目的 探讨甘露醇对大鼠局灶性脑缺血再灌注损伤脑组织NO及神经细胞凋亡的影响。方法 SD大鼠 4 8只 ,随机等分成 4组 ,每组 12只。A组 :假手术组 ;B组 :模型组 ,即局灶性脑缺血再灌注组 ;C组 :小剂量甘露醇组 ;D组 :大剂量甘露醇组。采用硝酸还原酶法及TUNEL法分别检测各组大鼠脑组织NO含量及神经细胞凋亡数目。结果 B组脑组织NO含量及神经细胞凋亡数目均明显多于A组 (P <0 .0 1) ;C组、D组脑组织NO含量及神经细胞凋亡数目均较B组减少 (P <0 .0 5 ) ;C组与D组脑组织NO含量及神经细胞凋亡数目比较 ,无显著差异 (P >0 .0 5 )。结论 甘露醇的脑保护作用不完全与脱水降颅压有关 ,还可能与下调缺血再灌注后脑组织NO含量、抑制神经细胞凋亡有关。Objective To investigate the effect of mannitol on NO and neural apoptosis in focal cerebral ischemia and reperfusion.Methods Forty-eight SD rats were randomly divided into four groups. Group A: sham-operated group(n=12);Group B: model group or ischemia and reperfusion group(n=12); Group C: treated by low dose of mannitol group(n=12); Group D: treated by high dose of mannitol group(n=12). The methods both nitrate redutase and TUNEL were respectively used to measure NO content and the number of neural apoptotic cells.Results NO content and the number of neural apoptotic cells in group B were higher than those in group A (P< 0.01). Compared with group B, NO content and the number of neural apoptotic cells were lower in group C and group D (P< 0.05). There was no siginifcant difference between group C and group D (P> 0.05).Conclusions The neuroprotective effects of mannitol are not completely related to its dehydration effect, but associated with decreasing NO content and inhibiting neural apoptosis.
