奥美拉唑对大鼠胃上皮细胞凋亡的影响

摘　　要：目的 :探讨奥美拉唑对大鼠胃上皮凋亡的影响。方法 :将 1 4只Wistar大鼠随机分为 2组 :实验组予以奥美拉唑 (40mg/kg)灌胃 1次 /d ,共 7d ;对照组以生理盐水代替。 7d后处死动物 ,分别取胃底、胃体、胃窦部黏膜标本 ,用TUNEL法检测胃上皮细胞凋亡情况。结果 :对照组、实验组胃底、胃窦、胃体部凋亡指数分别为 :4.98± 1 .64、8.1 2± 2 .61 ;4.69± 1 .58、7.2 9± 2 .34 ;3 .82± 1 .52、6 .68±2 .65 ,实验组与对照组相比 ,有显著性差异 (P <0 .0 5) ,两组组内比较无显著性差异 (P >0 .0 5)。结论 :奥美拉唑在体内能诱导大鼠胃上皮细胞凋亡 ,凋亡增加可能是过度增殖和突变的刺激因素 。Objective: To study the effect of omeprazole on the apoptosis of gastric epithelial cells in rats.Methods:14 Wistar rats were randomly divided into 2 groups:the experimental group was infused with omeprazole(40 mg/kg) intragastric ally once a day for 7 days,the control group was replaced with saline,the animals were killed 7 days after treatment.The tissue of the gastric fundus,corpus and antrum were taken to examine the apoptotic cells in situ using method of terminal uridine deoxynucleotidyl transferase mediated neck end labelling tchnique.Results:The apoptotic indexes of control group in the fundus,antrum and corpus were 4.98±1.64,4.69±1.58,3.82±1.52,respectively.While in the experimental group the indexes were 8.12±2.61,7.29±2.34,6.68±2.65,respectively.There was significantly difference between these 2 groups,but no significantly difference in the individual group.Conclusion:Omeprazole can induce gastric epithelium apoptosis in vivo.Increased apoptosis may be a stimulus for a compensatory hyperproliferation and potentially mutagenesis in normal mucosa,or a balance for the hyperproliferation.
