摘 要:目的:探讨大鼠心肌缺血损伤与凋亡蛋白Bcl-2和Bax的关系及牛磺酸的影响。方法:选择健康SD大鼠30只,随机分为假结扎组、结扎组和牛磺酸保护组,每组10只。结扎大鼠冠状动脉左前降支建立心肌缺血模型。检测3组心肌线粒体中超氧化物歧化酶(superoxidedismutase,SOD)、Ca2+-ATP酶活性和丙二醛含量,用免疫组化法检测心肌中的Bcl-2和Bax蛋白。结果:结扎冠状动脉左前降支可致心肌线粒体丙二醛含量升高犤假结扎组:(4.89±0.54)μmol/g;结扎组:(9.85±0.68)μmol/g犦,SOD和Ca2+-ATP酶活性下降犤假结扎组:(8.63±0.35)μmol/(g·s),(13.97±1.97)mmol/(g·s);结扎组:(6.73±0.39)μmol/(g·s),(8.54±2.28)mmol/(g·s)犦。缺血心肌Bax蛋白表达呈显著升高(t=3.931,P<0.01),牛磺酸能明显减少缺血心肌线粒体丙二醛的生成犤牛磺酸保护组:(5.46±0.72)μmol/g犦,降低心肌Bax蛋白的表达,增加Bcl-2蛋白的表达(t=3.716,P<0.01)。结论:结扎大鼠冠状动脉左前降支所致心肌缺血损伤与Bcl-2和Bax蛋白的表达有关,牛磺酸对缺血心肌Bcl-2和Bax蛋白的表达有较好的调控作用。AIM:To discuss the relationship between myocardium ischemia injury and apoptin Bcl 2 and Bax and the effects of taurine. METHODS:Thirty cases of healthy SD rats were selected and randomly allocated into pseudo ligation group,ligation group and taurine protection group with 10 cases in each group.The left descending branch of coronary artery of the rat was ligated to establish myocardium ischemic model.Superoxide dismutase(SOD),Ca2+ ATPase activity and malondialdehyde (MDA) content in myocardium mitochondrion were detected in 3 groups. Apoptin Bcl 2 and Bax were detected by immunohistochemical test. RESULTS:MDA contents in mitochondrion increased[pseudo ligation group (4.89± 0.54) μ mol/g;ligation group(9.85± 0.68) μ mol/g]and SOD and Ca2+ ATPase activity decreased[pseudo ligation group(8.63± 0.35) μ mol/(g· s),(13.97± 1.97) mmol/(g· s);ligation group(6.73± 0.39) μ mol/(g· s),(8.54± 2.28) mmol/(g· s)]due to the ligation of the left descending branch of coronary artery.The expression of Bax apoptin in ischemic myocardium significantly increased(t=3.716,P< 0.01).Taurine could significantly decrease MDA formation in mitochondrion[taurine protection group:(5.46± 0.72) μ mol/g]as well as the expression of Bax apoptin but increasing the expression of Bcl 2 apoptin(t=3.716,P< 0.01). CONCLUSION:There is a relationship between myocardium ischemic injury due to the ligation of the left descending branch of coronary artery and the expressions of apoptin Bax and Bcl 2.Taurine has a preferably regulative and controlling effect on the expression of Bax and Bcl 2 apoptins in ischemic myocardium.
