摘  要:目的 比较研究气虚证与热证模型大鼠鼻咽组织cDNA阵列A区基因表达谱特征及其对治疗的反应性。方法 采用药物加疲劳法和热性中药灌胃法分别制作大鼠气虚证和热证模型,并分组予针对性治疗。分别取实验动物鼻咽组织Atlas TM cDNA阵列法检测各组动物鼻咽上皮细胞基因组A区基因表达谱特征,分析其变化模式。结果气虚证模型动物表现c-jun、c-kit原癌基因表达下调,前胸腺素a和钙结合蛋白2基因表达上调;治疗一周后,表达上调的2基因活性恢复正常,但表达下调的2基因活性仍维持低表达状态。热证模型动物表达下调之基因同气虚证者,但表达活性上调者为绒毛蛋白、前胸腺素a、含铜锌超氧化物歧化酶1、热休克蛋白27和70基因;治疗一周后,所有表达上调之基因活性均恢复正常,但表达下调者活性尚未出现明显变化。结论 在细胞周期调节因子和应激反应蛋白基因活性表达上,气虚证和热证Atlas TM cDNA阵列A区基因表达谱特征有明显差异,但在癌基因表达活性方面显示出一致性。Objective To comparatively investigate the features of gene expression profiling in nasopharyngeal tissues on A area of Atlas?cDNA array between rats with Qi deficiency pattern and heat pattern respectively. Methods Qi deficiency pattern was induced with Senna leaf decoction perfused gastrically and supplemented by forced swimming in deep pool, and heat pattern was induced with the decoction made from heat herbs perfusing gastrically, all being prepared among rats. Then, they were randomly divided into modeling group (MG) and treating one (TG), with corresponding therapies against these patterns to treat the animals in TGs. By the end of therapy, animals were killed and nasopharyngeal tissues were taken from the nasopharynx for assay of gene expression profiling on A area of Atlas?cDNA array. After this, a comparative analysis was made to see the expressive features of genes on A area and their differences between the two groups before and after treatment. Results Among the animals with Qi deficiency, the expressive activities of c-jun and c-kit proto-oncogenes were down-regulated and those of prothy-mosin a and calcium binding protenin 2 genes were up-regulated. In contrast, up-regulated were the expressive activities of ezrin, prothymosin a, Cu-Zn SOD1, HSP27 and HSP70 genes among the animals with heat pattern, and down-regulated were the same genes as those in the animals with Qi deficiency. All the expressive activities of genes showing up-regulated were brought to the normal level while those showing down-regulated were still remained in a lowered level of expression following a course of therapy for one week. Conclusion There is a significant difference in the activity expression of genes responsible for cell cycle regulation and stress response protein transcription between the two groups of animals with different pattern, i. e. Qi deficiency or heat pattern, while it is almost the same in the expressive activity of proto-oncogenes for them.
