摘  要:目的 :探讨白介素 10 (IL 10 )、血小板衍生生长因子 BB (PDGF BB)对体外活化的培养大鼠肝星状细胞(HSC)表达细胞间黏附分子 1(ICAM 1)mRNA的影响。方法 :体外培养激活的HSC (细胞系rHSC 99)随机分为 4组 :对照组 (A组 )、IL 10 2 0ng/ml干预组 (B组 )、PDGF BB 2 0ng/ml干预组 (C组 )及IL 10 2 0ng/ml +PDGF BB2 0ng/ml共干预组 (D组 )。加药后 2 4小时 ,采用半定量逆转录聚合酶链反应 (RT PCR )方法 ,检测各组ICAM 1mRNA表达情况。结果 :B组ICAM 1mRNA的表达较A组明显降低 (P <0 0 1) ;C组ICAM 1mRNA的表达较A组明显增强 (P <0 0 1) ;D组ICAM 1mRNA的表达较A组与C组均降低 (P <0 0 5 ,P <0 0 1) ,但与B组相比较其ICAM 1mRNA的表达则增强 (P <0 0 1)。结论 :PDGF BB促进肝纤维化与其引起HSC表达ICAM 1增强有关 ,IL 10通过下调HSCICAM 1表达 ,可在抑制肝纤维化中发挥作用。Objective:To investigate the effects of IL-10 and PDGF-BB on the expression of ICAM-1 mRNA at activated rat hepatic stellate cells.Methods:Activated hepatic stellate cells were cultured and divided into 4 groups randomly: group A was contol group, group B was treated with IL-10 (20ng/ml),group C was treated with PDGF-BB(20ng/ml),group D was treated with IL-10(20ng/ml) and PDGF-BB(20ng/ml). The level of ICAM-1 mRNA was measured by the quantitative reverse-transcription polymerase chain reaction (RT-PCR) after the treatment for 24 hours. Results:The expression of ICAM-1 mRNA in group B was significantly decreased compared with that of group A(P<0.01). Whereas the level of group C was significantly increased(P<0.01). The level of group D was lower than that of group A and group(P<0.01),and higher than that of group B(P<0.01). Conclusion:The abrotic mechanism PDGF-BB partly due to its upregulation effects on ICAM-1. IL-10 had a antifibrotic effect on liver fibrosis through down-regulating the expression of ICAM-1.
