内皮抑制素治疗大鼠C6脑胶质瘤的实验研究

摘　　要：目的 :观察内皮抑制素对大鼠脑胶质瘤的治疗作用 ,并探讨治疗剂量的选择。方法 :建立大鼠脑胶质瘤模型后第 15天 ,以 5mg Kg·d、10mg Kg·d和 2 0mg Kg·d 3种剂量内皮抑制素皮下注射治疗 7d ,用磁共振成像观察肿瘤体积的变化。结果 :治疗结束时 ,3种剂量的肿瘤生长抑制率分别为 2 8.2 9± 3.4 1%、5 3.77± 7.0 8%和 5 9.4 6±3.2 0 % ,5mg剂量组与 10mg和 2 0mg组间有显著统计学差异 ,而 10mg组与 2 0mg组间差异无显著性。结论 :内皮抑制素对胶质瘤有治疗作用 ,治疗剂量应选择 10mg Kg·d。Objective:To evaluate the effect of endostatin upon rat brain glioma,and to probe the optimal therapeutic dosage.Methods:At the 15 th day after tumor implantation,SD rats were treated with different dosage of endostatin (5mg/Kg·d,10mg/Kg·d,20mg/Kg·d)and the treatment was maintained for 7 days.MRI was performed to observe the change of tumor volume.Results:The rates of tumor growth suppression in three groups were 28.29±3.41%,53.77±7.08% and 59.46±3.20% accordingly.The difference was significant between 5mg group and the other two groups,but there was no significant difference between 10mg group and 20mg group.Conclusion:Endostatin has therapeutical effects on glioma,and the effective dosage occurs in association with 10mg/Kg·d.
